摘要
杂交链式反应(Hybridization chain reaction,HCR)是一种新型的信号扩增技术,它利用分子识别和杂交反应依次打开多个发卡探针,得到累计信号,实现选择性检测靶分子的目的。其特点是整个反应过程无酶参与,避免了非特异性扩增对分析结果的影响,且反应条件温和,易控制。基于这些特点,HCR受到了研究人员广泛的关注,并结合各种DNA探针用于检测各种目标物,如:DNA、蛋白质、生物小分子、金属离子等等。该文总结了近些年来HCR反应在生物传感探针设计方面的应用。
Hybridization chain reaction (HCR) is a new type of signal amplification technology, based on a chain reaction of recognition and hybridization events between two sets of DNA hairpin molecules, offers an enzyme-free alternative for the rapid detection of specific targets. The advantage is that the whole reaction process without enzyme involved, avoiding the non-specific amplification of the analysis results. The reaction conditions are mild and easy to control. Combination with a variety of DNA probes, HCR based DNA analysis strategies are used for the detection of various targets, such as, DNA, proteins, small biomolecules, metal ions and so on. Here we review the biosensor probe design involving HCR in recent years.
