摘要
目的探究锁阳乙酸乙酯提取物(ECS)对去势后慢性应激小鼠认知功能障碍MAPK/ERK1/2信号通路中磷酸化的胞外信号调控激酶1/2(P-Erk1/2)和磷酸化的c AMP反应元件蛋白(P-CREB)表达量的影响。方法将实验动物采用随机数字法分为对照组、模型组、ECS组,除对照组外,模型组和ECS组,采用去势后慢性应激造模,ECS组同步灌胃ECS。在造模8周后采用Morris水迷宫检测小鼠空间学习记忆能力,造模成功时取材海马组织检测Western Blot。结果:Morris水迷宫结果显示,模型组较对照组穿越平台次数减少(P<0.01),ECS组较模型组穿越平台次数增加(P<0.01)。Western Blot结果显示,P-Erk1/2蛋白表达量模型组较对照组显著减少(P<0.01),ECS组较模型组显著增加(P<0.01);P-CREB蛋白表达量模型组较对照组显著减少(P<0.05),ECS组较模型组显著增加(P<0.05)。结论 ECS改善去势后慢性应激小鼠认知功能障碍机制之一可能是通过上调MAPK/ERK1/2信号通路中P-Erk1/2与P-CREB的蛋白表达量。
Objective To explore the impacts of the ethyl acetate extract of cynomorium songaricum(ECS) on the expressions of P-Erk1/2 and P-CREB of the MAPK/ERK1/2 signal pathway for the improvements in cognitive dysfunction in the mice of ovariectomized chronic stress. Methods According to the random number method,the experimental animals were divided into a control group,a model group and an ECS group. Except the control group,in the model group and ECS group,the ovariectomization was adopted to prepare the chronic stress model. Simultaneously,the gavage with ECS was applied in the ECS group. In the week 8 after modeling,Morris water maze test was used to determine the space learning and memory ability.When the modeling was successful,the hippocampal tissue was collected for the Western blotting test. Results The Morris water maze results showed that the frequency of passing-through platform in the model group was reduced as compared with the control group(P〈0. 01). It was increased in the ECS group as compared with the model group(P〈0. 01). The Western blotting result showed that the expression of P-Erk1/2 in the model group was reduced significantly as compared with that in the control group(P〈0. 01). It was increased significantly in the ECS group as compared with the model group(P〈0. 01). The expression of P-CREB was reduced significantly in the model group as compared with the control group(P〈0. 05) and it was increased in the ECS group as compared with the model group(P〈0. 05). Conclusion The up-regulation of P-Erk1/2 and P-CREB expressions in MARK/ERK1/2 signal pathway is possibly one of mechanisms of ECS on the improvements in the cognitive dysfunction in the mice of ovariectomized chronic stress.
