摘要
目的:研究黄芪甲苷对实验性糖尿病大鼠睾丸组织的保护作用并探讨其机制。方法:通过高脂高糖饮食加腹腔注射链脲佐菌素(STZ)的方法制备实验性糖尿病大鼠模型;测定大鼠空腹血糖水平,称量体质量和睾丸质量并计算睾丸指数,测定血清中睾酮含量,睾丸组织中酸性磷酸酶(ACP)、乳酸脱氢酶(LDH)和谷氨酰转肽酶(γ-GT)含量,测定睾丸组织中超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)、过氧化氢酶(CAT)活性和丙二醛(MDA)含量,HE染色观察睾丸组织形态结构,透射电镜观察睾丸组织超微结构。结果:黄芪甲苷能够显著降低实验性糖尿病大鼠空腹血糖水平,增高体质量、睾丸质量以及睾丸指数,升高血清睾酮含量,升高LDH、γ-GT、ACP含量和SOD、GSH-Px、CAT活性且降低MDA含量,显著改善睾丸组织结构病变和超微结构。结论:黄芪甲苷可能通过降低氧化应激反应、改善睾丸组织病变和细胞超微结构而对实验性糖尿病大鼠睾丸组织起到一定的保护作用。
Objective: To investigate the protective effects of astragalosides IV on testicular tissue of experimental diabetic rats, and explore its mechanism. Methods: Experimental diabetic rats were prepared by feeding them high fat, high sugar diet and intraperitoneal injection of STZ; the levels of fasting blood glucose was determined, the body weight and testicular mass were weighed, and testicular index was calculated, the contents of testosterone in the serum, the contens of ACP, LDH and γ-GT in the testicular tissue were determined, the activity of SOD, GSH-Px, CAT and the contents of MDA in the testicular tissue determined, morphological structure of testis ob served by HE staining, and the ultrastructure of testis tissue by transmission electron microscope (TEM). Results: Astragalosides IV could notably lower the levels of fasting blood glucose of experimental diabetic rats, increase body mass, testicular mass and testicular index, raise the contents of testosterone in the serum, the contents of ACP, LDH and γ -GT in the testicular tissue, the activity of SOD, GSH-Px, CAT and decrease the contents of MDA, as well as improve pathological changes and cell ultrastructure of testicular tissue notably. Conclusion: Astragalosides IV could protect testicular tissue of experimental diabetic rats to a certain degree, and it might be related to decreasing oxidative stress, improving pathological changes and cell ultrastructure of testicular tissue.
