摘要
目的通过检测骨桥蛋白(osteopontin,OPN)在膀胱移行细胞癌(transitional cell carcinoma of the bladder,TCCB)组织及癌旁正常膀胱组织中的表达情况,探讨其在肿瘤组织中的表达与肿瘤的临床分期、病理分级之间的关系,为临床病理诊断及预后判断提供一定的理论依据。方法收集2010年1月至2015年9月在本院泌尿外科手术切除的TCCB组织标本65例,应用免疫组织化学S-P法对65例TCCB组织以及10例癌旁正常膀胱组织标本进行OPN的检测,结合这些患者的临床病历资料进行分析:(1)OPN在TCCB组织与癌旁正常膀胱组织中的表达差异;(2)OPN在TCCB组织中的异常表达与TCCB的临床分期、病理分级之间的关系。结果 OPN在TCCB组织中的阳性表达率为47.7%,在癌旁膀胱正常组织中不表达,其阳性表达率与TCCB的病理分级和临床分期密切相关(P<0.05)。结论 OPN对TCCB的病理诊断有一定的意义,可能与TCCB的发生存在一定的关联,可作为TCCB病理诊断的标记物之一;同时,OPN可作为TCCB临床分期及病理分级的一项客观指标,可能与肿瘤的侵袭及转移密切相关,可以帮助判断患者的预后。
Objective To evaluate the expressions of osteopontin( OPN) in transitional cell carcinoma of the bladder( TCCB) and adjacent normal bladder tissue,to analyze the relations between the expression of OPN and clinical stage and pathological grade of the patients,in order to provide certain theoretical basis for pathological diagnosis and prognosis of TCCB. Materials Immunohistochemistry was employed to determine the expression of OPN in 65 cases of TCCB and 10 cases of adjacent normal bladder tissues collected from January 2010 to September 2015 in our department. The different expression levels of OPN in TCCB and adjacent normal bladder tissue were detected in the study. The relationship between the OPN expression and clinical stage and pathological grade of the patients was analyzed. Results The positive expression of OPN in cells is the distinctive brown stain. The positive rate of OPN expression in TCCB is higher than that in the adjacent normal bladder tissue.The positive expression closely associated with clinical stage and pathological grade of the patients( P < 0. 05).Conclusions OPN have a role in the development of TCCB,and the expression level of OPN should be employed to pathological diagnosis. Then,OPN should have a role in the tumor invasion and metastasis,and be used as an index of tumor stage,pathological classification and the prognosis of patients.
