蓝莓花色苷对大鼠脑缺血再灌注损伤的保护作用初探 被引量:4

Protective effect of blueberry anthocyanins on cerebral ischemia reperfusion injury in rats

摘要 目的观察蓝莓花色苷对大鼠脑缺血再灌注损伤的保护作用,初步探讨其作用机制。方法将100只SD大鼠随机分为5组,分别为假手术组、模型组和蓝莓花色苷给药低、中、高3个剂量组(50、100和200 mg/kg·d)。假手术组只分离右侧的颈总动脉、颈内动脉和颈外动脉,但不阻断血流。实验组采用线栓法建立大鼠脑缺血再灌注模型,观察蓝莓花色苷对脑缺血再灌注损伤大鼠的神经功能缺损评分、脑组织含水量的影响。取大鼠右侧脑组织,检测氧化物歧化酶(SOD)活性和丙二醛(MDA)含量,Elisa法检测脑组织中匀浆中核因子kappa B(NF-κB)、肿瘤坏死因子(TNF-α)、白细胞介素1(IL-1)和白细胞介素6(IL-6)水平,Western blotting法检测热休克蛋白70(HSP70)、磷酸化蛋白激酶B(p-Akt)表达水平。结果与模型组比较,蓝莓花色苷各剂量组可降低大鼠神经功能缺损评分(P<0.01),减少脑组织含水量(P<0.05),提高SOD活性,降低MDA含量(P<0.05),降低NF-κB、TNF-α、IL-1和IL-6水平(P<0.05);免疫印迹结果显示,蓝莓花色苷各剂量组均可增强p-Akt蛋白表达,其中中、高剂量组可明显增强HSP-70蛋白表达。结论蓝莓花色苷对大鼠脑缺血再灌注损伤具有较好的保护作用,其作用机制可能通过上调p-Akt和HSP-70蛋白的表达,发挥其对神经细胞的保护作用。 Objective To observe the protective effect of blueberry anthocyanins on the cerebral ischemia reperfusion injury in rats. Methods One hundred SD rats were randomly divided into 5 groups: the sham group, the model group, the low-dose, middle-dose and high-dose of blueberry anthocyanins groups(50, 100 and 200 mg/kg, d). In the sham group, only separatecommon carotid artery (CCA),internal carotid artery (ICA) and external carotid artery (ECA), but the blood flow was not blocked. The rat cerebral ischemia reperfnsion model was established by the middle cerebral artery occlusion method in rats, in order to observe the effect of blueberry anthocyanins on neurologic deficit score and the content of water in brain tissue. Right brain tissue of rats was taken to detect the content of SOD and MDA. The levels of NF-κB, TNF-α, IL-1 and IL-6 in the brain tissue were detected by Elisa. Western blotting method was used to detect the protein expression levels of HSP70 and p-Akt. Result Compared with the model group, the blueberry anthocyanins in each dose group could significantly decrease the neurologic deficit scores (P 〈 0.01 ) and the contents of water in brain tissue of the rats( P 〈 0.05 ), increase the activity of SOD and decrease the levels of MDA( P 〈 0.05 ) , decrease the levels of NF-κB,TNF-α.IL-1 and IL-6( P 〈 0.05 ), the result of Western blotting showed that three groups of blueberry anthocyanins could significantly increase the p-Akt protein expression, and the middle-dose and high-dose groups could increase HSP-70 protein expression. Conclusion Blueberry anthocyanins could protect the cerebral ischemia reperfusion injury of rats. Its mechanism may be related to up-regulated the expression of p-Akt and HSP-70 and play its protective role on neurons,
