摘要
目的探讨血管内皮生长因子(VEGF)和基质金属蛋白酶-9(MMP-9)与子痫前期发病的相关性。方法纳入江苏省常州市武进人民医院2013年1月-2016年11月确诊为子痫前期的患者100例,其中重度子痫前期患者49例,轻度子痫前期的患者51例,同时纳入同期分娩的健康孕妇50例作为对照组。采用Western blotting、半定量逆转录聚合酶链反应(RTPCR)分子生物学方法检测3组患者胎盘组织中VEGF、MMP-9的表达水平;采用酶联免疫吸附法(ELISA)检查患者血清及尿液中VEGF、MMP-9水平。结果 Western blotting、RT-PCR方法检测结果:轻、重度子痫前期组患者胎盘组织的VEGF、MMP-9表达水平均低于正常对照组,组间相比差异具有统计学意义(P<0.05);ELISA检测结果:两组血清及尿液中VEGF、MMP-9水平均低于正常对照组,且组间相比差异具有统计学意义(P<0.05)。结论 VEGF、MMP-9参与了子痫前期的病理生理改变,且与病情的发生及轻(重)程度有密切的相关性。
Objeetlve To explore the correlation between vascular endothelial growth factor （VEGF）, matrix metalloproteinase-9 （MMP-9） and preeelampsia. Methods From January 2013 to November 2016,100 pregnant women who were diagnosed preeclampsia in Wujin People Hospital were enrolled in the study, and 49 pregnant women were severe preeclampsia, 51 pregnant women were mild preeclampsia. At the same time, 50 healthy pregnant women were enrolled as control group. The levels of VEGF and MMP-9 in placenta were detected by Western blotting and RT-PCR. The levels of VEGF and MMP-9 in serum and urine were measured by enzyme-linked immunosorbent assay （ELISA）. Results Western blotting and RT-PCR showed that the expression levels of VEGF and MMP-9 in placenta of pregnant women with mild and severe preeclampsia were lower than those in control group, and the difference was statistically significant （ P 〈 0.05 ）. The levels of VEGF and MMP-9 in the serum and urine in pregnant women with mild and severe preeclampsia than those in control group, and the difference was statistically significant （ P 〈 0.05 ）. Conclusion VEGF and MMP-9 are involved in the pathophysiological changes of preeelampsia and are closely related to the occurrence and severity of preeclampsia.
