摘要
目的建立胰岛素抵抗细胞模型,探讨岩藻黄素体外改善胰岛素抵抗及降血糖活性。方法采用高浓度胰岛素诱导HepG2细胞,筛选最佳诱导条件建立IR-HepG2细胞模型。用不同浓度岩藻黄素处理模型细胞,用葡萄糖氧化酶法(GOD-POD)检测岩藻黄素对IR-HepG2细胞葡萄糖消耗量的影响。结果 10-6 mol/L胰岛素处理细胞24h为建立IR-HepG2细胞模型的最佳条件。5~20μmol/L的岩藻黄素均可促进IRHepG2细胞对葡萄糖的利用,10、12.5和15μmol/L组的糖消耗量显著高于模型组(P<0.01)。结论岩藻黄素可改善体外IR-HepG2细胞的胰岛素抵抗,促进IR-HepG2细胞的葡萄糖消耗,具有改善胰岛素抵抗和降血糖活性。
Objective To establish insulin-resistant HepG2 cell model and study the effect of fucoxanthin on improving the insulin resistance and the hypoglycemic activity in vitro.Methods High concentrations of insulin were used to induce the insulin resistance in HepG2 Cells.The best concentration and induction time of insulin were selected to establish the insulin-resistant HepG2 Cell model(IR-HepG2).IRHepG2 cells were treated with different concentrations of fucoxanthin.The glucose consumption of different groups was tested by glucose oxidase-peroxidase method(GOD-POD).Results The best concentration and treatment time of insulin to induce insulin resistance in HepG2 cells were 10-6 mol/L and 24 h.Results of the glucose consumption test showed that fucoxanthin in the concentration range of 5~20μmol/L could stimulate the glucose utilization of the IR-HepG2 cells.Glucose consumption in the groups of 10,12.5 and 15μmol/L of fucoxanthin were all significantly higher than those of the IRHepG2 model group(P〈0.01).Conclusion Fucoxanthin could improve the insulin resistance and promote the glucose consumption of IR-HepG2 cells in vitro.Fucoxanthin has hypoglycemic activity.
