摘要
采用离子液体作为反相高效液相色谱流动相添加剂,建立了同时测定莲子心中莲心碱、异莲心碱和甲基莲心碱含量的方法.色谱柱为SPHERIGEL ODS C18(250 mm×4.6 mm I.D.,5μm),流动相为添加30mmol/L[HMIM][BF_4]、pH为3.0的甲醇/水(70/30,v/v),流动相流速为1.0mL/min,检测波长为280nm.在浓度范围为2-200μg/mL范围内,莲心碱、异莲心碱和甲基莲心碱的线性相关系数分别为0.999 8,0.999 7和0.999 8;对莲心碱、异莲心碱和甲基莲心碱的LOD分别为0.14 mg/L、0.17 mg/L和0.19 mg/L,LOQ分别为0.29 mg/L、0.35mg/L和0.41mg/L.
A reversed phase high performance liquid chromatography(RP-HPLC)method for simultaneous determination of liensinine,isoliensinine and neferine in Nelumbinis Plumula was established by using 1-hexyl-3-methylimidazolium tertafluoroborate([HMIM][BF_4])as the mobile phase additives.A SPHERIGEL ODS C18 column(250 mm×4.6 mm I.D.,5μm was used for separation.Methanol/water(70/30,v/v)containing 30 mmol/L [HMIM][BF4]was used as the mobile phase.The pH of the mobile phase was 3.0 and the flow rate of the mobile phase was 1.0 mL/min.The detection wavelength was 280 nm.In the range of 2-200μg/mL,the linear correlation coefficients were all better than 0.999 7.For liensinine,isoliensinine and neferine,the LOD of this method was 0.14,0.17 and 0.19 mg/L,respectively,and the LOQ of this method was 0.29,0.35 and 0.41 mg/L,respectively.
