星点设计-效应面法优化纤维素酶法辅助提取鬼针草叶中总黄酮研究 被引量:4

摘要 目的:优化纤维素酶法辅助提取鬼针草叶中总黄酮工艺。方法:在单因素考察基础上选取酶用量、酶解pH值、酶解温度三个影响因素,采用星点设计-效应面法优化纤维素酶辅助提取过程中的影响因素及其交互作用对鬼针草叶中总黄酮提取率的影响,建立二项式拟合模型。结果:最佳提取工艺为酶用量1.14%,酶解温度41.9℃,酶解pH值4.35,该条件下总黄酮提取率为6.31%,与模型预测值相差仅为1.25%。结论:星点设计-效应面法适用于纤维素酶法辅助提取鬼针草叶中总黄酮提取工艺的优化,所建立的数学模型预测性良好。 Objective:To extract total flavonoids from Bidens pilosa Linn. leaves through optimized cellulose process. Methods:Based on single-factor experiments,three factors of the enzyme amount,pH value,enzymolysis temperature were chosen to optimize the cellulase assisted extraction total flavonoids from Bidens pilosa Linn. leaves by central composite design and response surface method. Results:The best extraction process was as follows:the enzyme amount 1.14%,enzymolysis temperature 41.9 ℃,pH value 4.35;the optimized extraction rate of total flavonoids was 6.31%,which was different from the model prediction by 1.25%. Conclusion:Central composite design and response surface method was suitable for the extraction process optimization of cellulose assisted extraction total flavonoids from Bidens pilosa Linn. leaves. The established mathematical model for the predictability was satisfactory.
