摘要
目的:探讨山慈菇水煎剂对人乳腺癌T-47D细胞增殖、迁移的影响。方法:利用细胞培养技术对人乳腺癌T-47D细胞进行培养;利用MTT法比较不同浓度山慈菇水煎剂组和对照组对T-47D细胞生长增殖情况的影响;利用细胞划痕实验,检测不同浓度的山慈菇水煎剂对T-47D细胞迁移的影响。结果:不同浓度的山慈菇水煎剂(50 g·L^(-1)、100 g·L^(-1)、200 g·L^(-1),400 g·L^(-1)、600 g·L^(-1)、800 g·L^(-1))对T-47D细胞的生长抑制率分别为-0.60%、-17.10%、-0.60%、65.08%、81.15%、82.80%,400~800 g·L^(-1)质量浓度山慈菇水煎剂对乳腺癌T-47D细胞的增殖有明显抑制作用;划痕实验结果显示200 g·L^(-1)、300g·L^(-1)山慈菇水煎剂作用24h和48h后,与对照组比较,乳腺癌T-47D细胞的划痕距离较大,划痕未见明显愈合,差异有统计学意义(P<0.05)。结论:一定浓度的山慈菇水煎剂对T-47D细胞生长增殖具有抑制作用,低浓度(<200 g·L^(-1))的山慈菇水煎剂对T-47D细胞的生长增殖无影响,即无细胞毒性;高浓度(>200 g·L^(-1))的山慈菇水煎剂对乳腺癌T-47D的增殖有明显的抑制作用;一定浓度的山慈菇水煎剂可以抑制T-47D细胞的迁移运动。
Objective: To investigate the effects of Iphigenia decoction on proliferation and migration of human breast cancer T-47 D cell. Methods: Human breast cancer T-47 D cell were cultured by using cell culture technique. MTT method was used to compare the growth situation of T-47 D cells in treatment group and control group. Cell migration in T-47 D cells was evaluated by using scratch test. Results: Iphigenia decoction with 50 g·L-1,100 g·L-1,200 g·L-1,400 g·L-1,600 g·L-1,800 g·L-1could inhibit the T-47 D cells proliferation. The inhibition rate was-0. 60% and-17. 10%,-0. 60%,65. 08%,81. 15% and82. 80% respectively. The results of the scratch test indicated that T-47 D cells treated by 200 g·L-1,300 g·L-1Iphigenia decoction for 24 h and 48 h had significantly lower wound healing rate compared with that of the control group( P 0. 05). Conclusion: Low concentrations( 200 g·L-1) Iphigenia decoction had no effect on T-47 D cells proliferation. High concentration( 200 g·L-1) Iphigenia decoction had obvious inhibitory effects on breast cancer T-47 D proliferation. Certain concentration Iphigenia decoction can inhibit T-47 D cells migration.
