摘要
目的探讨分析苦瓜总皂苷对顺铂所致卵巢损伤大鼠是否有治疗作用,并分析作用机理。方法 60只雌性SD大鼠适应性喂养2周后随机分为正常组、模型组、苦瓜总皂苷低、中、高剂量组以及阳性药物逍遥丸组,每组10只,造模成功后给予对应药物治疗6周后处死大鼠,收集血清和卵巢组织,测定SOD、CAT、MDA、凋亡蛋白cytc、卵泡刺激素(FSH)及雌二醇(E2)含量;同时免疫组化、Western和半定量PCR检测卵巢组织中PPARγ表达。结果与正常组相比,模型组血清中cytc和FSH含量明显增高(P<0.05),E2含量明显降低(P<0.05),苦瓜总皂苷可明显降低cytc和FSH(P<0.05),升高E2(P<0.05),但逍遥丸对此3个指标却没有作用;与正常组相比,模型组血清和卵巢组织中SOD以及CAT含量明显降低(P<0.05),MDA含量明显升高(P<0.05),苦瓜总皂苷和逍遥丸均可以升高SOD和CAT(P<0.05),并降低MDA(P<0.05),此外随着苦瓜总皂苷给药剂量的增加疗效更为显著;与正常组相比,模型组PPARγ表达明显降低(P<0.05),苦瓜总皂苷能明显增加PPARγ表达量(P<0.05),且效果明显好于逍遥丸组。结论苦瓜总皂苷能显著改善顺铂所致卵巢组织氧化损伤,且呈剂量依赖性,这可能与其可激活PPARγ有关。
Objective To observe the effect of total momordicoside to the ovarian injury in rats caused by cisplatin,and analyze its mechanism.Methods After 2 weeks adaptive feed,60 female SD rats were randomly divided the normal group,model group,total momordicoside low,middle,high dose groups and xiaoyao pill group(n=10),when the rats model were successful built and given the corresponding drug treatment,after 6 weeks,the rats were sacrificed,collected the serum and ovarian tissue to observe the super oxide dismutase(SOD),catalase(CAT),malondialdehyde(MDA),serum cytc,follicle-stimulating hormone(FSH) and serum estradiol(E2) levels; finally,immunohistochemical,Western and semi-quantitative PCR were used to detect the PPARγ expression in ovarian tissue.Results Compared with the normal group,model group decreased significantly in the SOD and CAT(P0.05),MDA increased significantly(P0.05),total momordicoside and Xiaoyao pill can obviously increase the content of SOD and CAT(P0.05),and decrease MDA(P0.05); for the Cytc,compared with normal group,model group was significantly higher(P0.05),and FSH levels were significantly lower in the E2(P0.05),the total momordicoside can obviously reduce cytc and FSH levels(P0.05) and increase E2(P0.05); as for PPARγ,compared with the normal group,model group has significantly lower expression(P0.05),total momordicoside can significantly increase expression,and its curative effect was better than the Xiaoyao pill group(P0.05).Conclusion Total momordicoside can significantly improve the ovarian tissue oxidative damage caused by cisplatin,and it shows obvious concentration dependence,which is related to the expression of PPARγ.
