摘要
目的检测p66Shc在心力衰竭大鼠血清中的表达并探讨其与氧化应激的关系。方法SD大鼠分为心力衰竭组(HF组)和正常对照组(对照组)。检测心力衰竭大鼠血清超氧化物歧化酶(SOD)的活力及丙二醛(MDA)的含量;超声心动图检测心室舒张末内径(LVEDD)、左心室收缩末内径(LVESD)、左室射血分数(LVEF);RT-PCR检测血p66ShcmRNA的表达,并对p66Shc的表达与LVEDD、LVESD、LVEF、MDA、SOD进行相关分析。结果心力衰竭组SOD、LVEF明显下降,MDA、p66Shc明显升高,差异有统计学意义(P〈0.05);心力衰竭组p66ShcmRNA的表达量与LVEDD、LVESD、MDA呈正相关(r=0.636、0.620、0.645,P〈0.05),而与LVEF、SOD呈负相关(r=-0.860、-0.650,P〈0.05)。结论p66Shc与心力衰竭氧化应激密切相关,p66shc可能参与心力衰竭氧化应激的过程,可为心力衰竭新的治疗途径提供依据。
Objective To investigate the expression of adaptor protein p66shc in rats with heart failure and its relationship with oxidative stress. Methods Rat heart failure model was established by partially banding abdominal aorta.The levels of serum superoxide dismutase ( SOD ) and malondialdehyde ( MDA ) activity were examined.The left ventricular end diastolic diameter ( LVEDD ) , left ventricular end systolic diameter ( LVESD ) , left ventricular ejection fraction ( LVEF ) were measured by routine cardiac ultrasonography.The expression of p66Shc was determined by quantitative real time PCR. Results The levels of SOD and LVEF were significantly lower while the levels of MDA and p66Shc were significantly higher in HF group than that in control group ( P〈0.05 ) .Linear correlation analysis showed that the expression of gene p66Shc was positively related with the level of LVEDD, LVESD and MDA ( P〈0.05 ) , but negative related with the level of LVEF and SOD ( P〈0.05 ) . Conclusion P66shc is associated with states of increased oxidative stress. P66shc mRNA levels are increased in HF, which may be involved in the pathogenesis of HF through oxidative stress.
