摘要
核糖体DNA内转录间隔区(internal transcribed spacer,ITS)是被子植物分子系统发育研究中应用最广泛的分子标记。对收集的12份安徽地方特色桑种质资源提取总DNA,用PCR扩增方法进行测序分析;根据鲁桑基因序列,确定ITS序列范围,并进行序列比对;通过分析序列长度、G+C含量、变异位点及遗传分歧,探讨了安徽桑树种质材料间的亲缘关系,为更深入地研究桑属植物间的系统进化关系提供重要的参考资料。
Ribosome transcribed spacer(ITS)is the most widely used nuclear DNA sequence in molecular systems. The sequencing analysis was carried out by using the total DNA extraction and PCR amplification method. According to the sequence of Morus multicaulis gene, the ITS sequence range was determined and sequence alignment was performed. By analyzing the sequence length, G + C content and mutation loci and genetic divergence, the phylogenetic relationship between the collected mulberry germplasm in Anhui Province was discussed, which provided the basic reference for the further study of phylogenetic evolution between mulberry plants.
