摘要
目的:研究豚鼠感染呼吸道合胞病毒(RSV)后不同时间内肺组织M胆碱能受体数量和功能的改变。方法:建立RSV感染豚鼠的动物模型,用放射配基结合分析法测定肺组织M胆碱能受体的数量和亲和力。结果:鼻内滴入RSV3天后,豚鼠肺组织就出现炎症改变,5~7天达高峰,14天基本恢复正常。肺组织病毒分离及免疫荧光检查均证实为RSV感染。放射配基结合分析表明肺组织M受体在感染RSV后3天就出现受体最大结合容量(Bmax)增高,平衡解离常数(Kd)值降低,第5、7天时,Bmax达高峰,Kd值达最低值,第21天仍与对照组差异显著(P<0.05)。结论:鼻内滴入RSV可形成豚鼠RSV感染模型。RSV感染后引起肺组织M受体数量的增加,亲和力上升,这种改变持续时间较长,是引起气道高反应性的主要原因之一。
Objective:To investigate the effects of respiratory syncytial virus(RSV)infection on M receptor density and function in lung tissue of guinea pig.Methods:After intranasal inoculation of guinea pig with RSV,the virus rapidly proliferated in lung tissue.By using the method of specific binding of isotopic ligand to M receptors the quantity and affinity of M receptor in lung tissues were measured on days 3,5,7,9,11,14 and 21 after inoculation.Results:The RSV induced group showed pathological evidence of acult bronchiolitis 3rd day after inoculation,which reached the peak during the days 5 to 7 and recovered on day 14.RSV infection had significant effect on the dissociation constant(Kd)as well as the maximum binding capacity(Bmax)of M receptor.The Kd appears decreased on day 3,which reached the lowest on day 5 to 7 and was still low on day 21.Bmax appears increased on day 3,which reached the peak on day 5 to 7 and was still high on day 21.Conclusion:A model of acult RSV bronchiolitis was prepared by intranasal inoculation of human RSV into guinea pigs.The RSV infection may increase the affinity and density of M receptor which may play a role in airway hyperreactivity.
