摘要
目的探讨血浆中miR-21、miR-155的表达水平在胰腺癌诊断中的价值。方法选择32例胰腺癌患者作为胰腺癌组,选择性别、年龄相匹配的27例健康体检正常者作为对照组,采集2组血液样本及临床病理数据,采用RT-PCR法检测血浆中miR-21、miR-155的表达量,采用电化学方法检测同样本中的CA19-9含量,分析miR-21、miR-155表达量与胰腺癌患者临床参数的关系,评估miR-21、miR-155及CA19-9诊断胰腺癌的效能。结果胰腺癌组血浆中miR-21、miR-155表达量和CA19-9含量均明显高于对照组(P均<0.05);胰腺癌患者血浆miR-21、miR-155表达量和肿瘤临床分期呈正相关(P<0.05),与其他参数无明显相关性;miR-21、miR-155联合CA19-9检测诊断胰腺癌的灵敏度和特异度均明显高于单项检测(P均<0.05)。结论血浆中miR-21、miR-155检测有助于胰腺癌的早期诊断。
Objective It is to investigate the clinical value of plasma microRNAs detection in diagnosis of pancreatic carcinoma.Methods 32 cases of pancreatic carcinoma patients were selected to pancreatic carcinoma group,and 27 cases of ageand-gender-matched physical examination for healthy people to control group,plasma samples and complete clinical data were collected from the two groups,and the the levels of miR-21 and miR-155 in the plasma were detected by quantitive reverse transcription polymerase chain reaction (qRT-PCR),and the content of CA19-9 were detected by electro-chemiluminescence (ECL).The relationship of miR-21 and miR-155 expression with clinical characteristics in pancreatic cancer patients were analyzed,and the diagnostic efficiency of miR-21,miR-155 and CA19-9 in pancreatic carcinoma.Results The expression of miR-21 and miR-155 and the content of CA19-9 in the pancreatic carcinoma group were increased compared to the control group (all P〈0.05).The plasma miR-21 and miR-155 level showed a positive correlation with TNM stage in pancreatic cancer patients (all P〈0.05).The sensitivity and specificity of combined detection of miR-21 or miR-155 and CA19-9 in the diagnosis of pancreatic cancer were significantly higher than those of the two individual detection (all P〈0.05).Conclusion The detection of miR-21 and miR-155 in plasma may provide a new method in the early diagnosis of pancreatic cancer.
