摘要
目的初步探讨缺氧时Pept1、Oatp1b1、Oat1、Oct1、Mdr1和Mrp2 6种药物转运体基因表达的变化。方法Wistar大鼠随机分为正常对照组、缺氧24h和72h组,缺氧组置于模拟海拔5000m(压力:397.5mmHg,氧分压:42mmHg)的低压氧舱中。提取各组大鼠小肠、肝脏、肾脏组织中的总RNA,以Real-time PCR方法检测6种药物转运体基因表达量。结果与正常对照组相比,缺氧组中6种药物转运体基因表达大多数升高,但随着时间的延长,不同组织中的不同转运体出现不一样的变化趋势。结论高原缺氧会影响药物转运体mRNA的表达水平,这可为高原药物转运体及药代动力学研究提供一定的理论依据。
Objective To study the gene expressions of six drug transporters-Peptl, Oatplbl, Oatl, Octl,Mdrl and Mrp2 in rats after acute hypoxia at high altitude. Methods Wistar rats were randomly divided into normal control group, hypoxia 24 h group and hypoxia 72 h group. The experimental groups (hypoxia 24 h and 72 h groups) were fed in a hypobaric chamber simulated to an altitude of 5000 m (pressure: 397.5 mmHg, the oxygen partial pressure: 42 mmHg). Total RNA was extracted from the small intestine, liver and kidney of rats in each group. Real-time PCR was used to detect the expression levels of six transporter genes.Results Compared with control group, the expressions of most of the drug transporters Peptl, Oatplbl, Oat1, Octl, Mdrl and Mrp2 in hypoxia groups were increased, but differ- ent transporters in different tissues developed diverse trends with the extension of hypoxia time.Conclusion Hypoxia at high altitude can affect the mRNA expression levels of drug transporters, which may provide some reference for the study of drug transporters and pharmacokinetics at high altitude.
