摘要
目的探究高强度超声联合微泡开放血脑屏障对大鼠脑组织的影响及形态学变化特点。方法将SD大鼠15只随机分为3组(对照组、微泡组、微泡+超声照射组),每组5只。对照组大鼠尾静脉注射生理盐水2mL,不用超声照射;微泡组大鼠注射造影剂0.2mL,尾静脉注射生理盐水2mL,不用超声照射;微泡+超声照射组大鼠注射造影剂0.2mL,尾静脉注射生理盐水2mL,用超声频率12 MHz的探头(机械指数设置为1.9)照射大鼠脑部颞窗,并用伊文思蓝作为追踪剂,肉眼观察各组大鼠伊文思蓝染色情况,荧光显微镜观察伊文思蓝荧光红斑情况,光镜观察各组大鼠脑组织的形态学变化。结果肉眼观察对照组和微泡组大鼠脑组织未见伊文思蓝染色,光镜观察未见脑组织水肿及形态学变化,荧光显微镜观察脑组织少见红色荧光光斑。肉眼观察微泡+超声照射组大鼠脑组织可见伊文思蓝染色,荧光显微镜观察脑组织可见多个卵圆形红色光斑,光镜观察可见脑组织水肿,红细胞渗出,个别细胞坏死。结论高强度超声联合造影剂能够成功开放大鼠血脑屏障,同时对大鼠脑组织有一定程度的损伤。
Objective To investigate the effect of high intensity ultrasound combined with microbubbles on the blood-brain barrier(BBB)as well as on its morphological changes of rats.Methods A total of 15 SD rats were randomly divided into 3 groups(control group,microbubble group,microbubble with ultrasonic irradiation group),with each group having 5rats.The control group whose rats were given intravenous injection of saline 2 mL without ultrasound irradiation;microbubble contrast agent group whose rats were injected with 0.2 mL intravenous injection of saline 2 mL without ultrasound irradiation;microbubbles with ultrasound irradiation group whose rats were injected with contrast agent 0.2mL,intravenous injection of saline 2mL with ultrasonic probe the frequency of 12MHz(mechanical index set to 1.9)irradiation in rat brain and temporal window,with Evans blue being as a tracer.the morphological changes was observed by the naked eye using Evans blue staining,erythema was observed with Evans blue by fluorescence microscopy,the morphological changes of brain tissue of rats in each group was observed using light microscopy.Results NO morphological changes and brain edema can be observed of rats in control group and microbubble group by Evans blue staining,and few red florescent spot was found,either;Evans blue staining can be visible using naked eyes and many oval red spots were seen under the fluorescent scope,brain edema and extravasation of brain tissues were present under the light microscope,with several cell being necrosis in microbubbles with ultrasonic group.Conclusion High intensity of ultrasound combined with contrast agents was shown to be able to successfully open the BBB of rats and damage the brain tissue of rats to some extent.
