摘要
目的探讨GOLPH3基因对宫颈癌细胞Hela迁移和侵袭能力的影响。方法通过脂质体转染法将GOLPH3的表达质粒和靶向GOLPH3的siRNA转入Hela细胞内;通过荧光实时定量PCR检测GOLPH3基因的相对表达量,Transwell实验检测Hela细胞迁移和侵袭能力。结果荧光实时定量PCR结果显示,与NC组(0.990±0.005)相比,siRNA组GOLPH3表达量(0.383±0.020)显著降低(P<0.01),与pcDNA组(0.997±0.029)相比,GOLPH3组GOLPH3表达量(3.333±0.219)显著升高(P<0.01)。Transwell细胞迁移实验结果显示,NC、siRNA、pcDNA和GOLPH3组具有迁移能力的细胞数分别为(172.30±4.33)、(75.00±3.79)、(163.70±3.93)和(258.00±4.93)。Transwell细胞侵袭实验结果显示,NC、siRNA、pcDNA和GOLPH3组具有侵袭能力的细胞数分别为(53.67±2.96)、(26.67±2.73)、(51.67±2.19)和(81.67±3.38)。结论上调GOLPH3基因表达可促进宫颈癌细胞迁移和侵袭能力;反之,抑制其表达可抑制宫颈癌细胞迁移和侵袭能力。
Objective To investigate the effect of GOLPH3 on migration and invasion of cervical cancer cell Hela. Methods GOLPH3 expression plasmids and siRNAs were transfected into Hela cells using lipofactamine. The mRNA expression of GOLPH3 was examined by quantitative real-time RT-PCR (qRT-PCR). The migration and invasion abilities of Hela cells were evaluated using Transwell assays. Results The results of qRT-PCR showed that the expression level of Group siRNA (0.383 ±0.020, n=3) was significantly lower than that of Group NC 0.990±0.005 (P〈0.0 1), and the expression level of Group GOLPH3 3.333 ± 0.219 was significantly higher than that of Group pcDNA (0.997±0.029) (P〈0.01). The results of Transwellmigration assay showed that the number of migrated cells in Group NC, siRNA, pcDNA and GOLPH3 was 172.30 ±4.33, 75.00± 3.79, 163.70 ± 3.93 and 258.00 ±4.93, respectively. The results of Transwellinvasion assay showed that the number of invasive cells in Group NC, siRNA, pcDNA and GOLPH3 was 53.67±2.96, 26.67±2.73, 51.67±2.19 and 81.67±3.38, respectively. Conclusion The upregulated expression of GOLPH3 enhances the migration and invasion abilities of cervical cancer cells. Conversely, inhibiting the expression can inhibit the migration and invasion abilities of cervical cancer cells.
