摘要
研究顺铂对大鼠血浆、胃及孤束核(NTS)ghrelin及其受体表达的影响。将大鼠分为顺铂组和生理盐水组。记录摄食、高岭土摄入量和体重。48h后分别取血分离血浆,采用Elisa法测定ghrelin浓度;取胃组织冷冻保存,采用PCR法测定ghrelin及其受体GHS-R1a mRNA的表达;取NTS冷冻保存,采用Western blot和PCR法测定ghrelin和ghrelin受体GHS-R1a蛋白及其mRNA表达水平。顺铂组与生理盐水组比较,食物摄入量减少(p<0.05);高岭土消耗量显著增加(p<0.01);体重减少但无统计学意义(p>0.05);血浆ghrelin浓度降低但无统计学意义(p>0.05);胃ghrelin mRNA表达下降,但无显著意义(p>0.05),受体GHS-R1amRNA表达显著增加(p<0.05);NTS ghrelin蛋白及其mRNA表达下降(p<0.05),受体GHS-R1a蛋白及其mRNA表达显著增加(p<0.05)。ghrelin可能参与顺铂引起的恶心呕吐。
To study the effect of cisplatin on the expression of ghrelin and its receptors in plasma,stomach and nucleus of solitary tract(NTS)in rats.The rats were divided into the cisplatin group and the saline group.The feeding,kaolin intake and body weight were recorded.After 48 h,blood plasma was separated,and ghrelin concentration was determined by Elisa method;Cryopreservation of gastric tissue,the expression of ghrelin and its receptor mRNA GHS-R1 awas determined by PCR method.Cryopreservation of NTS,ghrelin and ghrelin receptor GHS-R1 aprotein and mRNA expression levels were determined by Western blot and PCR methods respectively.Compared with saline group,the food intake in cisplatin group decreased(p〈0.05);kaolin consumption was increased significantly(p〈0.01);body weight decreased with no statistical significance(p〉0.05);plasma ghrelin concentration decreased but no statistical significance(p〉0.05);gastric mRNA expression of ghrelin decreased,but there was no significant meaning(p〉0.05),while mRNA expression of ghrelin receptor GHS-R1 asignificantly increased(p〉0.05);mRNA expression of ghrelin in NTS decreased as well as its protein expression(p〈0.05),while ghrelin receptor GHS-R1 a expression significantly increased(p〈0.05).Ghrelin may be involved in cisplatin induced nausea and vomiting.
