摘要
分别以去离子水、乙醇和丙酮作为沉淀剂,研究了不同沉淀剂对离子液体(1-丁基-3-甲基咪唑醋酸([BMIM]Ac))中再生胶原的三股螺旋结构和细胞相容性的影响。结果表明:去离子水再生胶原的红外光谱(FT-IR)中I_(1240/1450)值最大(0.97),圆二色谱(CD)的正负吸收峰的强度比值(Rpn)最高(0.148),差示扫描量热仪(VP-DSC)的吸热峰值(Td)达38.3℃,3种结构表征方法均表明去离子水再生胶原的三股螺旋结构的保留度最高,而丙酮再生胶原最低;原子力显微镜(AFM)观察发现:3种再生胶原都显示出了互相交织成网的纤维结构,表明[BMIM]Ac溶解胶原的过程并没有破坏胶原的三股螺旋结构,但丙酮再生的纤维结构较为稀疏凌乱;细胞相容性试验表明:3种再生胶原均具有促进成纤维细胞增殖的能力,而去离子水再生胶原细胞的相容性最好,成纤维细胞的纺锤形最为典型,表明去离子水是离子液体[BMIM]Ac中胶原再生的最佳溶剂。
Deionized water, ethanol and acetone were used as precipitators, the effect of different precipitators on the triple helical structure and cytocompatibility of collagen regenerated from ionic Iiquid ( [ BMIM ] Ac) was investigated. The results indicate that the value of I1240/1450(0. 97) in Fourier transform infrared spectrometer (FT-IR), the ratio of positive and negative absorption peak intensity (Rpn, 0. 148) in circular dichroism spectrum (CD) together with the denature temperature (Td, 38.3℃ ) in differential scanning calorimeter (VP-DSC) of collagen regenerated from deionized water all exhibit the highest values, revealing that the triple helical structure of collagen regenerated from deionized water is well remained and superior to the collagen regenerated from acetone. Under observation of a- tomic force microscope ( AFM), three types of regenerated collagens all exhibit intertwined fiber network map, which confirms that the dissolving process of collagen by [ BMIM ] Ac did not destroy the triple helix structure of collagen, but acetone has negative effect on intrinsic structure of collagen, so the regenerated fiber from acetone displays more sparse and messy fibrous structure. Cytocompatibility experiments show that three kinds of regenerated collagen all can improve the fibroblast cell reproduction, while the collagen regenerated from deionized water has the best compatibility and most typically spindle shape of fibroblast cells, indicating that the deionized water is the best candidate for the re- generation of collagen in ionic liquid [BMIM] Ac.
