黄芪汤通过AMPK-Camkkβ(LKB1)信号通路对足细胞的保护作用研究 被引量:4

摘要 目的通过体外实验研究,探讨AMPK-Camkkβ(LKB1)信号通路在黄芪汤改善糖尿病肾病中的作用。方法以高糖刺激足细胞为模型,黄芪汤进行干预。采用CCK8检测高糖状态下黄芪汤对足细胞存活率的影响。通过蛋白质免疫印迹法(Western blot)检测各组足细胞AMPK、p-AMPK、Camkkβ、p-Camkkβ、LKB1、p-LKB1、ACC、p-ACC蛋白的表达量。结果黄芪汤可浓度依赖性的改善高糖刺激下足细胞的活性,并浓度依赖性的上调高糖刺激下足细胞中p-AMPK、p-Camkkβ、p-LKB1、p-ACC蛋白表达水平,从而激活Ⅱ型糖尿病肾病诱导的AMPK-Camkkβ(LKB1)信号通路下调。结论黄芪汤可有效活化AMPK-Camkkβ(LKB1)信号通路,改善Ⅱ型糖尿病模型诱导的糖尿病肾病。 Objective To investigate the role of AMPK-Camkkβ(LKB1) signaling pathway in improving the diabetic nephropathy by Huangqi decoction by in vitro experimental study.Methods With high glucose to stimulate podocytes as a model,Huangqi decoction to intervene.The effect of Huangqi decoction on viability of podocytes was detected by CCK8.The expression levels of AMPK,p-AMPK,Camkkβ,p-Camkkβ,LKB1,p-LKB1,ACC,p-ACC protein were detected by western blotting.Results The concentration of p-AMPK,p-Camkkβ,p-LKB1,p-ACC protein in podocytes stimulated by high glucose was increased in a concentration-dependent manner,and the activity of paclitaxel was increased in a concentration-dependent manner.Activation of AMPK-Camkkβ(LKB1) signaling pathway induced by type 2 diabetic nephropathy.Conclusion Huangqi decoction can effectively activate AMPKCamkkβ(LKB1) signaling pathway to improve diabetic nephropathy induced by type 2 diabetes mellitus.
