摘要
目的:利用PCR Array技术观察健脾祛痰化瘀中药对脾虚痰浊动脉粥样硬化(AS)巴马小型猪主动脉AS信号通路相关基因表达变化的影响,探讨健脾祛痰化瘀方防治AS的可能机制。方法:将15只巴马小型猪随机分为正常组、模型组和治疗组,每组5只,正常组普通饲料喂养,模型与治疗组给予高热量高脂饲料喂养并进行冠脉球囊挤压伴拉伤及跑步措施进行干预至24周,制备巴马小型猪病症结合脾虚痰浊AS模型,治疗组除上述造模措施外给予健脾祛痰化瘀方灌胃24周。呼吸机麻醉后取材,提取各组主动脉总RNA,应用AS信号通路PCR芯片检测健脾祛痰化瘀方对脾虚痰浊AS巴马小型猪主动脉AS信号通路相关基因mRNA表达变化的影响。结果:PCR Array结果显示:与空白组相比,模型组显著上调的基因共13个,如ACE、BAX、SELE等基因,显著下调的基因共16个,如BCL2A1、LDLR、LPL等基因;与模型组相比,治疗组显著上调的基因共14个,如BCL2A1、LDLR、BCL2等,显著下调的基因共13个如BAX、PTGS1、SOD1等。结论:健脾祛痰化瘀方可能参与调控主动脉AS信号通路中脂质代谢与凋亡等相关基因表达从而起到防治脾虚痰浊AS巴马小型猪AS形成及稳点斑块的作用。
Objective : By using PCR array technique to observe the effect of Jianpi Qutan Huayu Decoction on spleen deficienc and phlegm turbid syndrome Bama miniature pig aorta atherosclerotic signaling pathway related genes expression and explore the possible mechanism of Jianpi Qutan Huayu Chinese medicine in the prevention and treatment of atherosclerosis. Methods: Fifteen Bama miniature pigs were randomly divided into normal group, model group and treatment group,5 in each group. The normal group was fed with normal diet. The model and treatment groups were given high calorie high - fat diet and coronary balloon compression with strain and running measures of intervention for 24 weeks to prepare the Bama mini pigs with spleen deficiency and phlegm turbid syndrome of atherosclerosis. The treatment group in addition to the above modeling measures was given Jianpi Qutan Huayu Decoction by gavage. After ventilator anesthesia, total RNA was extracted from each group' s aorta and use signaling pathway PCR chip to detect the influence of Jianpi Qutan Huayu Decoction on Bama miniature pig' s atherosclerotic signaling pathway related genes mRNA expression. Results:PCR showed compared with the blank group ,the model group's significantly upregnlated genes were 13, such as ACE, BAX,SELE;significantly down regulated genes were 16, such as BCL2A1 ,LDLR, LPL. And compared with the model group, treatment group' s significantly upregulated genes were 14, such as BCL2A1, LDLR,BCL2 ;significantly down regulated genes were 13, such as BAX, PTGS1 ,SOD1 and so on. Conclusion: Jianpi Qutan Huayu Decoction may be involved in the regulation of aortic atherosclerosis signaling pathway in lipid metabolism and apoptosis related gene expression and control spleen deficiency and phlegm turbid syndrome Bama miniature pig' s atherosclerosis formation and steady plaque.
