摘要
建立了一种测定卡托普利的双波长褪色分光光度法。在pH 5.76的Tris-盐酸缓冲溶液中,卡托普利与孔雀石绿反应生成具有两个明显负吸收峰的绿色离子缔合物,最大负吸收波长位于634 nm,次大负吸收波长位于588 nm,表观摩尔吸光系数(κ)分别为1.35×10~4和1.05×10~4[L/(mol·cm)],当用双波长褪色法测定时,表观摩尔吸光系数(κ)达2.40×10~4[L/(mol·cm)],线性范围为0~3.9(mg·L^(-1)),服从朗伯-比尔定律。还优化了反应条件,探讨了吸收光谱特征和共存物质的影响。方法用于市售卡托普利药物中卡托普利的测定,结果满意。
A new double-wavelength fading spectrophotometric method for the determination of captopril was established. In Tris-hy- drochloric acid buffer solution of pH 5. 76,captopril reacted with malachite green to form a green ion association complexes with ob-vious two negative absorption peak. The maximum negative absorption wavelength located at 634 nm and the second largest negative absorption wavelength located at 588 nm. Their apparent molar absorptivity(/c) were 1. 35 × 10^4 an d 1. 05 × 10^4 [ L / ( m o l · c m ) ] respectively. When the double-wavelength fading spectrophotometric method was used to determine the concentrations of captopril, its apparent molar absorptivity( k ) was 2. 40×10^4 [ L/( mol · cm) ] an d linear range of 0 -3. 9 ( m g ·L ^- 1) an d captopril obeys L a m - bert-Beer's law in definite mass concentration range of 0 ?3. 9 ( mg · L^- 1 ). Also optimized the reaction conditions,a nd the absorp-tion spectral characteristics and the effects of coexisting substances were discussed. The method was applied to determine the con-tent of captopril in commercially available captopril drugs with satisfactory results.
