摘要
目的探讨电针干预对大鼠脑出血后同侧海马区Slit2表达的影响及其作用机制。方法将55只成年SD大鼠随机分为对照组和脑出血组,脑出血组采用Ⅶ型胶原酶脑内注射建立大鼠尾状核出血模型,脑出血组再随机分为非干预组和电针组。电针组于造模次日始行电针干预,非干预组不予任何干预。各组分别于术后24 h、7 d、14 d及21 d,随机取5只大鼠,取出血侧海马区脑组织,石蜡切片行免疫组化Slit2检测。结果脑出血组右侧尾状核区域有一直径约3 mm的血肿,光镜下见血肿区神经元变性坏死,大量红细胞渗出,血肿边缘有较多中性粒细胞与小胶质细胞浸润。对照组大鼠可见右侧海马区散在分布少量Slit2阳性细胞,而脑出血组Slit2阳性细胞数明显增多;较之对照组,各时间点非干预组、电针组Slit2阳性细胞数均显著增多(P﹤0.01),而电针组又明显多于非干预组(P﹤0.01)。结论电针干预可显著增强脑出血大鼠脑内Slit2的表达水平,有利于脑出血后中枢神经轴突的修复与再生,促进神经系统功能恢复。
Objective To explore the impacts of electro-acupuncture intervention on the Slit2 expression in ipsilateral hippocampus in rats following intracerebral hemorrhage and its mechanism of action. MeAods A total of 55 adult SD rats were randomly divided into a control group and a hemorrhagic group. The hemorrhagic group received type VII collagenase injection in the brain to establish a model of rat caudate nuclear hemorrhage and was randomly divided into a non-intervening group and an electro-acupuncture group. The electro-acupuncture group started electro-acupuncture intervention from the next day after modeling. The non-intervening group received no intervention. Five rats were randomly selected from each group 24 hours, seven days, 14 days and 21days after the operation to get the paraffin section of the brain tissue of the haemorrhagic hippocampus for immunohistochemical Slit2 detection. Results There was a hematoma with a diameter of about 3 mm in the right caudate nucleus area in the hemorrhagic group. Neuronal degeneration necrosis and the diapedesis of lots of erythrocyte were observed under light microscope in the hematoma region. There were many neutrophils and microglia cells infiltrated at the edge of the hematoma. A small number of Slit2 positive cells were observed in the right hippocampus in rats in the control group, while the number of Slit2 positive cells in the hemorrhagic group increased significantly; compared with the control group, the number of Slit2 positive cells at different time points in the non-intervening group and the electro-acupuncture group increased significantly (P 〈 0.01), and the number of Slit2 positive cells in the electro-acupuncture group was much more than that in the non-intervening group (P 〈 0.01). Conclusion Electro-acupuncture intervention can significantly increase the expression level of Slit2 in rats with cerebral hemorrhage, help the restoration and regeneration of central axons after cerebral hemorrhage and promote the recovery of nervous s
