摘要
目的:研究LPS诱导的炎症反应对脑动脉瘤形成和破裂引起出血的作用。方法:通过肾性高血压大鼠脑动脉瘤模型法建立大鼠动脉瘤模型进行如下处理:正常大鼠:A组注射生理盐水;B组注射LPS;大鼠动脉瘤模型鼠:C组注射生理盐水;D注射LPS。RT-PCR的方法检测动脉炎症因子NF-κB、MCP-1、IL1、IL6、TNF-α的表达水平。组织化学的方法检测SMA染色检测动脉壁的厚度,Ig染色检测动脉瘤破裂后出血面积。结果:LPS诱导动脉组织中炎症因子NF-κB、MCP-1、IL1、IL6、TNF-α的表达水平明显增加。LPS能够导致动脉瘤破裂的增多,出血量增加。通过A组和B组比较LPS本身虽然能够明显诱导炎症反应但并没有明显的影响脑动脉瘤形成和破裂引起出血。结论:LPS诱导的炎症反应,炎症细胞的血管浸润进一步加剧脑动脉瘤形成和破裂引起的出血。
Objective:Objective to study the effect of LPS induced inflammatory response on hemorrhage caused by cerebral aneurysm formation and rupture.Methods:By the method of rat model of cerebral aneurysm in renal hypertensive rat models of aneurysms were treated as follows:normal rats:A group were injected with saline injection group;B LPS;rat aneurysm model in rats:group C injection of saline injection;D LPS.RT-PCR method was used to detect the expression levels of NF-κB,MCP-1,IL1,IL6,and TNF-αin arterial inflammatory factor B.SMA staining was used to detect the thickness of arterial wall and Ig staining was used to detect the bleeding area after aneurysm rupture.Results:The expression of NF-κB,MCP-1,IL1,IL6,and TNF-αin the arterial tissue of rats induced by LPS was significantly increased by B.LPS increase the number of aneurysms and increase the amount of bleeding.Compared with A group and B group,LPS itself can induce inflammatory reaction,but it has no obvious influence on the formation and rupture of cerebral aneurysm.Conclusion:The inflammatory reaction induced by LPS and the infiltration of inflammatory cells further aggravate the hemorrhage caused by cerebral aneurysm formation and rupture.
