摘要
目的观察养肝解毒散结中药对肝癌肿瘤生长和细胞凋亡、细胞衰老的作用。方法采取裸小鼠皮下移植人肝癌Bel-7402细胞方法建立模型,然后随机将小鼠分为中药组和对照组各10只,分别给予养肝解毒散结中药和蒸馏水灌胃,每天1次。灌胃3周后,剥离肿瘤组织称质量,TUNEL法检测细胞凋亡情况,衰老相关β-半乳糖苷酶染色检测细胞衰老情况。结果中药组肿瘤质量明显低于对照组(P<0.05),细胞凋亡和细胞衰老光密度值均明显高于对照组(P均<0.05)。结论养肝解毒散结中药可以抑制肝癌肿瘤生长,其机制与促肝癌细胞凋亡和细胞衰老相关。
Objective It is to observe the effects of Yanggan Jiedu Sangjie （YGJDSJ） herbs on tumor growth, apoptosis and cell senescence in human hepatocellular carcinoma. Methods Nude mice subcutaneously transplanted with human hepato- carcinoma Bel-7402 cells were randomly divided into two groups （10 mice per group） , and intragastric administered with dis- tilled water in the control group and YGJDSJ herbs daily in the traditional Chinese medicine group. After three weeks of treat- ment, the tumors were dissected and weighed. Apoptosis were detected by TUNEL assay. Cell senescence was identified by Senescence-associated ~3-galactosidase （SA-^-gal） staining. Results The weight of tumor of traditional Chinese medicine group was significant lower than that of control group （P 〈 0.05） , and the optical density values of apoptosis and cell senes- cence were significantly higher than those of the control group （all P 〈 0.05 ）. Conclusion YGJDSJ herbs can inhibit hepatocarcinoma growth, and may relate to induction of apoptosis and cell senescence.
