摘要
观察二烯丙基二硫(DADS)对IL-1β诱导SD大鼠关节软骨细胞的影响,除空白对照组外,其余各组加入IL-1β(10 ng/m L)诱导24 h后换DADS处理,通过CCK8实验、平板克隆实验检测增殖能力,Western blot实验检测MMP13、TIMP1和CollagenⅡ蛋白表达水平。结果发现:12.5和25μmol/L DADS对IL-1β诱导SD大鼠关节软骨细胞有增殖作用(P<0.05),50和100μmol/L DADS对IL-1β诱导SD大鼠关节软骨细胞有抑制作用(P<0.05);12.5和25μmol/L DADS显著增加IL-1β诱导SD大鼠关节软骨细胞克隆集落形成能力(P<0.05),50μmol/L DADS显著抑制IL-1β诱导SD大鼠关节软骨细胞克隆集落形成能力(P<0.05);DADS能显著抑制MMP13的表达,增加TIMP1和CollagenⅡ的表达。以上结果提示DADS对IL-1β诱导SD大鼠关节软骨细胞具有双向调节作用,25μmol/L DADS保护关节软骨细胞的分子机制可能与DADS显著抑制MMP13的表达,增加TIMP1和CollagenⅡ的表达相关。
To explore the effect of diallyl disulfide (DADS) on articular cartilage cell induced by IL-1β in SD rats. The growth inhibition of articular cartilage cell induced by IL-1β in SD rats following DADS treatment was measured by CCK8 assay and plate colony formation assay; The protein expression of M M P 1 3 , T I M P 1 and Collagen II were deter-mined using Western blot. DADS (12. 5 and 25 jjimol/L) significantly increased the cell proliferation and high colony formation capacity; DADS (50 and 100 jjimol/L) significantly suppressed the cell proliferation and lowered colony for-mation capacity ( P 〈 0. 05 ) ; Western blot demonstrated DADS markedly down-regulated the expression of MMP-1β , and increased the expression of Collagen II and TIMP1 in the cells. These results indicated that DADS had two-way regula-tion effect on SD rat articular cartilage cell induced by IL-1β,DADS (25 jjimol/L) had a protective effect on SD rat ar-ticular cartilage cell induced by IL-1/3,DADS (50 and 100 jjimol/L) can inhibit articular cartilage cell by IL-1β in SD rats,the protection molecular mechanism was possibly related to down-regulation of the expression of MMP-1β by DADS, and increased the expression of Collagen II and TIMP1 on SD rat articular cartilage cell with osteoarthritis.
