紫铆花素对人黑素瘤细胞A375生物活性及线粒体膜电位的影响 被引量:4

摘要 目的观察紫铆花素对人黑素瘤细胞A375细胞增殖、细胞周期、酪氨酸酶活性、黑素合成及线粒体膜电位的影响。方法用不同浓度紫铆花素处理人黑素瘤细胞A375 48h后,采用MTT法测定黑素细胞增殖率;流式细胞仪测定黑素细胞周期和线粒体膜电位变化;L-DOPA法测定黑素细胞酪氨酸酶活性及Na OH裂解法测定黑素含量变化。结果与对照组相比,0.1~5.0μg/m L浓度范围紫铆花素与人黑素瘤细胞A375共孵育48h后,能明显促进人黑素瘤细胞A375增殖,具有不同程度提高酪氨酸酶活性、促进黑素合成及调节细胞周期和线粒体膜电位的作用。结论紫铆花素对体外培养的人黑素瘤细胞A375起促进细胞增殖、提高酪氨酸酶活性、增加黑素合成的作用,其作用可能与紫铆花素对细胞周期和线粒体膜电位的调节有关。 Objective To evaluate the effects of butein on the proliferation, cell cycle, tyrosinase activity, melanin synthesis and mitochondrial membrane potential of human melanoma cell line A375.Methods Human melanoma cell line A375 was treated with different concentrations of butein for 48h.Methyl thiazolyl tetrazolium(MTT)assay was performed to evaluate cell proliferation rate; flow cytometry was to evaluate cell cycle and mitochondrial membrane potential; dopa oxidation assay was to evaluate tyrosinase activity; a sodium hydroxide solubilization method was used to measure melanin content.Results Compared with untreated control group, human melanoma cell line A375 treated with butein in a concentration of 0.1~5.0μg/mL for 48h were all significantly improved proliferation rate,melanogenesis, tyrosinase activity and regulated cell cycle and mitochondrial membrane potentialin melanocytes.Conclusion The butein could upregulate proliferation rate,as well as tyrosinase activity,melanin synthesis.Therefore, the regulation of cell cycle and mitochondrial membrane potential may affect the biological activity of human melanoma cell line A375.
