牵牛子化学拆分组分对大鼠阿霉素肾病模型药效学研究 被引量:8

摘要 目的:观察牵牛子水煎液及拆分组分对大鼠阿霉素肾病药效学研究,找寻牵牛子对肾病模型作用的物质基础。方法:采用尾静脉注射阿霉素的方法建立大鼠阿霉素肾病模型。考察牵牛子水煎液及拆分组分对阿霉素肾病模型大鼠血清生化指标及IL-1β和TNF-α的影响。结果:与空白组比较,模型组模型组白蛋白、总蛋白含量明显降低;总胆固醇、总甘油三酯、血肌酐和尿素氮的含量明显升高;IL-1β、TNF-α和尿中蛋白含量明显升高;肾小球上皮细胞肿胀明显,肾小球毛细血管淤血并部分闭塞,系膜基质重度增生呈弥漫性分布。与模型组比较,水煎液0.63g/kg、多糖0.37g/kg和酚酸0.13g/kg可以升高白蛋白、总蛋白、血肌酐、尿素氮含量;可以降低总甘油三酯、总胆固醇含量;可以降低IL-1β和TNF-α的浓度;可以降低尿中蛋白浓度。光镜检查结果显示牵牛子水煎液0.63g/kg、多糖0.37g/kg和酚酸0.13g/kg能减轻上皮细胞肿胀及肾小球系膜基质增生,改善大鼠肾脏的病理变化。结论:牵牛子水煎液0.63g/kg、多糖0.37g/kg和酚酸0.13g/kg可以提高肾小球的滤过功能,调节血浆胶体渗透压的平衡及血脂紊乱,从而改善肾病综合征表症。作用机制可能通过降低一系列炎性细胞因子的含量,从而达到缓解水肿的疗效。 Objective: To study the pharmacodynamics of the decoction and its components on the adriamycin nephropathy in rats, and to find out the material basis of the effect of bovine kidney on the model of nephropathy. Methods : Adriamycin nephropathy model was established by injecting adriamyein into tail vein. To investigate the effect of Decoction and its components on serum biochemical indexes, IL-1 beta and TNF- alpha in rats with adriamyein induced nephropathy. Results: Compared with the blank group, model group, model group, albumin, total protein content decreased significantly; the content of total cholesterol, triglyceride, serum creatinine and blood urea nitrogen increased significantly; significantly increased the protein content of IL-1 beta, TNF- alpha and urine; glomerular epithelial cell swelling significantly, glo- merular capillary congestion and partial occlusion, severe hyperplasia of mesangial matrix diffuse distribution. Compared with the model group, the decoction of 0.63g/kg, polysaccharide 0.37g/kg and salvianolic acid 0. 13g/kg can increase albumin, total protein, serum creatinine, urea nitrogen content; can reduce the total triglyceride, total cholesterol; can reduce the concentration of IL-1 and TNF- alpha beta; can reduce the urinary protein concentration. The results of light microscopy showed that 0.63g/kg, 0.37g/kg and 0.13g/kg could reduce the swelling of epithelial cells and the proliferation of mesangial matrix, and improve the pathological changes of kidney in rats. Conclusion: The decoction, polysaccharide and phenolic acid components can improve the glomerular filtration function, regulate the balance of plasma colloid osmotic pressure and blood lipid disorder, and improve the symptoms of nephrotic syndrome. The mechanism of action may be to reduce the effect of a series of inflammatory cytokines to relieve edema.
