摘要
针对干眼症临床治疗对泪点栓产品的研发需求,文中利用壳聚糖(CS)与β-甘油磷酸钠(GP)可在37℃发生温敏相变形成凝胶的特性,采用生物相容性优良的海藻酸钠(SA)为共混材料,改进CS-GP凝胶存在温敏相变时间较长、机械强度差、易于溶胀和降解、热稳定性差等问题,并采用扫描电子显微测试、红外光谱分析、热重分析、压缩弹性模量和流变性能分析等方法对复合凝胶性能进行了表征。结果表明:制备的复合溶胶在37℃环境可快速相变形成具有多孔网络结构且强度适宜的凝胶,可注射性良好。当CS质量浓度为25 mg/mL,GP质量浓度为600 mg/mL,V(CS)∶V(GP)=4.5∶1时,添加40 mg/mL SA可将CS-GP-SA复合凝胶的相变时间调控为10 s以内,压缩弹性模量为67 k Pa,损耗模量G″和储能模量G'值比为0.003 1,平衡溶胀率为5%,置于模拟泪液中48 d时的质量残留率为61%。凝胶性能可以满足泪道栓塞的应用需求。
Aiming at developing punctual plug for clinical treatment of dry eye, a kind of composite thermo-sensitive hydrogel was prepared at 37 ℃ using chitosan (CS) and sodium glycerophosphate ( GP), especially blended with sodium alginate (SA). Sodium alginate was considered to overcome drawbacks of CS-CP hrdrogel such as long gelating time, weak mechanical strength, excessive swelling and degradation and poor thermal stability, etc.. The composite gels were characterized by scanning electron microscopy, infrared spectroscopy, thermogravimetric analysis, compressive elastic modulus and rheological properties. Under experimental conditions including 25 mg/ mL CS, 600 mg/mL GP, 40 mg/mL SA and volume ratio of V( CS): V(GP) =4.5: 1, the prepared composite can be rapidly changed from sol into gel at 37 ℃ within 10 s with a porous network structure as well as suitable mechanical strength. The compressive elastic modulus of the composite hydrogel was 67 kPa whereas the ratio of loss modulus G" to storage modulus G' was 0. 003 1. The swelling rate of composite hydrogel was 5% , and after being treated by stimulated tears for 48 d, the mass residual ratio remained 61%. Therefore, the prepared composite hydrogel possesses satisfied injectability to meet the application of punctal plug.
