Ikaros家族成员在造血系统发育过程中的作用与意义

摘要 造血细胞在发育过程中受许多转录因子调控,其中Ikaros家族是这些转录因子的重要代表之一。Ikaros转录因子具有特征性的保守结构模序,能够与含关键基因启动子或增强子的DNA特异性短序列结合,调控它们的转录活性。同时,Ikaros转录因子还和其它的相关转录调节子之间相互作用,共同调控造血细胞的发育和分化。从结构上观察Ikaros家族,它们属于锌指结构转录因子,包括Ikaros、Helios、Aiolos、Eos和Pegasus 5个成员,分别由IKZF-1、2、3、4、5基因编码。它们是造血作用的主要调节子,通过单独或共同调控发挥作用,特别对淋巴细胞等造血细胞的发生、发展以及功能的正常发挥中具有重要作用,家族成员功能异常时常与疾病的发生、发展相关。本文综述Ikaros家族近年来取得的研究成果,一方面有助于深入理解这个家族在造血系统中的作用及意义,另一方面也为进一步开展研究工作提供可能的研究方向。 Hematopoietic cells are regulated by many transcriptional factors during their development,among them the Ikaros family is one of the most important representatives. They have characteristic conserved structural motifs,binding with DNA specific short sequences-containing key gene promoter or enhancer,to regulate their transcription activity. Meanwhile,the Ikaros family interact with other related transcriptional regulators to regulate the development and differentiation of hematopoietic cells. The structure of the Ikaros family,which belong to the zinc finger transcription factor,including Ikaros,Helios,Aiolos,Eos and Pegasus,are encoded by IKZF1-5 genes,respectively. They are master regulators of hematopoiesis,playing important roles in the occurrence,development and function of hematopoietic cells such as lymphocytes via individual and joint regulation. When working abnormalities,they are often related with the occurrence and development of the disease. In this review,the research achievements of the Ikaros family in recent years are summarized. On the one hand,it is helpful to understand the role and significance of this family in hematopoietic system; on the other hand,it provides the possible research direction for further research work.
