摘要
目的:分析不同剂量的骨蚀宁胶囊对激素性股骨头坏死(INFH)动物模型蛋白-2(BMP-2)表达的影响,进一步探讨骨蚀宁胶囊治疗INFH的作用机理。方法:选取成年健康新西兰大白兔56只,随机选取6只为正常对照组,剩余50只用于骨坏死造模;造模成功后观察2周,将50只模型动物随机分为5组:骨蚀宁胶囊低剂量组、骨蚀宁胶囊中剂量组、骨蚀宁胶囊高剂量组、空白对照组和阳性对照组,每组10只;继续培养8周,比较6组动物BMP-2表达水平以及血栓素B2(TXB2)、6-酮-前列腺素F1α(6-keto-PGF1α)等指标。结果:TXB2水平比较为空白对照组>低剂量组>高剂量组>阳性对照组>中剂量组>正常对照组;6-keto-PGF1α水平比较为正常对照组>中剂量组、阳性对照组>高剂量组>低剂量组>空白对照组;BMP-2表达情况为正常对照组>中剂量组>高剂量组>低剂量组、阳性对照组>空白对照组,差异比较有统计学意义(均P<0.05)。结论:骨蚀宁胶囊可以显著改善兔股骨头局部组织瘀血状态,提高BMP-2表达水平,且中剂量组(1.41g/kg·d)用药效果比其他剂量组更佳。
Objective: To investigate the effect of different doses of Gushining capsule on the expression of protein-2(BMP-2) in steroid-induced femoral head necrosis (INFH), and to further explore the mechanism of the treatment of INFH. Methods: 56 adult New Zealand white rabbits were selected and randomly selected 6 as the normal control group,the remaining 50 for bone necrosis modeling; 2 weeks after modeling success, 50 model animals were randomly divided into 5 groups: low-dose group, Middle dose group, high dose group of Gushining capsule, the control group and positive control group, 10 rats in each group. The levels of BMP-2 and the levels of thromboxane B2 (TXB2) and 6-ketoprostaglandin F1α (6-keto-PGF1α) were compared after 8weeks. Results: TXB2 expression: blank control group〉low dose group〉 high dose group〉 positive control group〉 middle dose group〉 normal control group; 6-keto-PGF1 α expression: normal control group〉 middle dose group, positive control group〉 high dose group〉 low dose group〉 blank control group; BMP-2 expression: normal control group〉 middle dose group〉 high dose group〉 low dose group, positive control group〉 blank control group. The difference was statistically significant ( P〈0.05 ). Conclusion: Gushining capsule can significantly improve the blood stasis status of the femoral head in rabbits and improve the expression of BMP-2, and the effect of the middle dose group (1.41g / kg·d) is better than that of other dosage groups
