摘要
目的探讨2型糖尿病患者脂蛋白酯酶基因多态性与血甘油三酯水平之间关系。方法选取2014年2月—2016年2月该院收治的110例2型糖尿病患者以及抽取体检人群中110名健康体检者为研究对象,其中110例糖尿病患者为研究组,110名健康体检者为对照组。分别对两组人群进行抽血,从血液中提取DNA、PCR扩增LPL基因特定片段以及限制性内切酶,对两组人群的酶切产物进行分析。结果研究组患者的TG(1.1±0.3)mmol/L、TC(3.0±1.1)mmol/L、Apo B(1.1±0.2)mmol/L、FBG(3.9±1.0)mmol/L、Hb Alc(5.4±0.1)mmol/L以及HOMA-IR(1.5±0.9)mmol/L指标均高于对照组人群,而HDL-C(1.5±0.6)mmol/L指标低于对照组人群,两组差异有统计学意义(P<0.05)。研究组患者的G等位基因与CG基因型频率均低于对照组,差异有统计学意义(P<0.05)。研究组和对照组研究人员的LPL基因PvuⅡ酶切位点基因频率差异无统计学意义(P>0.05)。结论脂蛋白酯酶基因多态性与2型糖尿病患者无明显关联,但Ser447Ter突变会在一定程度上降低患者血甘油三酯水平,从而可能会降低2型糖尿病患者发病率。
Objective To investigate the relationship between lipoprotein lipase gene polymorphism and serum triglyceride levels in patients with type 2 diabetes mellitus. Methods A total of February 2014 to February 2016 in our hospital from 110 cases of type 2 diabetic patients and collected for the physical examination crowd in 110 cases of healthy persons as the research object, which diabetic patients as study group, healthy who as the control group. Blood samples were drawn from two groups of people. DNA and PCR were extracted from the blood to amplify the LPL gene specific fragment and restriction enzyme. The enzyme products of the two groups were analyzed. Results Patients in the study group, TG(1.1±0.3)mmol/L, TC(3.0±1.1)mmol/L, apoB (1.1±0.2)mmol/L, FBG(3.9±1.0)mmol/L, HbAlc (5.4±0.1) mmol/L and HOMA-IR(1.5±0.9) mmol/L index were higher than those of the control group, and HDL-C(1.5±0.6) mmol/L index below the control group; the difference between the two groups has statistical significance(P〈0.05).The frequencies of G allele and CG genotype were lower in the study group than in the control group, and the difference was statistically significant(P〈0.05). In the study group and control group, there was no significant difference in the frequency of Pvu II restriction sites of LPL gene(P〉0.05). Conclusion There was no obvious correlation lipid proteinesterase gene polymorphism and type 2 diabetes, but Ser447Ter mutation can reduce blood triglyceride levels in patients in a certain extent, which may reduce the rate of onset of type 2 diabetes.
