摘要
目的观察HOTAIR对胶质瘤细胞侵袭和转移能力的影响。方法使用q-PCR从临床标本中检测HOTAIR与E-cadherin的关系,体外常规培养SWO-38和U251细胞,过表达HOTAIR后使用划痕实验检测细胞侵袭、迁移能力;Western blotting检测细胞中E-cadherin蛋白的表达。结果 q-PCR胶质瘤组织中HOTAIR高表达组中E-cadherin表达的均值较低,较HOTAIR低表达组低,差异有统计学意义(P<0.05);在SWO-38和U251细胞中过表达HOTAIR后Western blotting检测发现E-cadherin表达减少,划痕实验显示过表达HOTAIR后细胞迁移能力增强。结论 HOTAIR可增加胶质瘤的侵袭和转移,可能与其降低E-cadherin蛋白的表达有关。
Objective To observe the effect of HOTAIR on invasion and metastasis of glioma.Methods Using q-PCR to test the relationship between HOTAIR and E-cadherin from clinical specimens.The invasion and metastasis of cells were tested through Wound Healing after HOTAIR overexperssion in the conventional culture SWO-38 and U251 cells in vitro.Western blotting was used to test the expression of C-cadherin protein in cells.Results In q-PCR glioma tissue,the expression average of Ecadherin was lower in HOTAIR high expression than that in HOTAIR low expression.The difference was statistically significant.After overexpression HOTAIR in SWO-38 and U251 cells,the expression E-cadherin was decreased through Western blotting detection.Wound Healing showed that the ability of metastasis was enhanced after HOTAIR overexpression.Conclusion HOTAIR can increase the invasion and metastasis of glioma,which may be related to the decreased E-cadherin expression.
