Objective: To compare the effects of procaine and bupivacaine under different temperature and different concentration on the excitability of the bullfrog sciatic nerve. Methods: 144 sciatic nerve from 72 bullfrog were dissected and randomly divided into 3 groups with 48 nerves in each group. The temperature of the drug is 18℃ for Group I, 25℃ for Group II, and 35℃ for Group III. Each group was divided into 4 sub-group. 1% and 0.5% procaine and bupivacaine were used to block the sciatic nerve for each sub-group, respectively. The action potential was recorded by a multi-channel physiological signal recorder. The onset time and recorvery time of the drug under different conditions were compared. Results: In the condition of same temperature and same concentration, the onset time of Procaine was significantly slower than that of bupivacaine. However, the recorvery time of Procain after removing the drug was much shorter than that ofbupivacaine. For the same drug, the onset time of 1% concentration was faster than 0.5% at the same temperature and the recorvery time of 1% was longer than that of 0.5%. The onset time was also affected by the temperature. The onset time was reduced with the increase of the temperature. But the recovery time was not affected by the temperature. Conclusion: Both temperature and concenlration could affect the oneset of the local anesthetics. Bupivacaine has shorter oneset time and longer recovery time than procaine at the same concentration.
