松叶牡丹离体快速繁殖及瓶苗微型化培育技术研究

摘要 应用植物离体快繁技术对松叶牡丹瓶苗微型化培育技术的研究,结果表明:松叶牡丹的种子用0.1%的氯化汞灭菌5min效果最理想,成活率达到73.3%;最佳的增殖培养基配方:MS+6-BA 1.0mg·L^(-1)+IBA 0.05mg·L^(-1)+蔗糖30g·L^(-1)+琼脂9g·L^(-1);对松叶牡丹的壮苗情况来看,PP333的最佳质量浓度为0.5mg·L^(-1),芽苗在的培养基为MS+NAA 0.25mg·L^(-1)+PP3330.5mg·L^(-1)+蔗糖30g·L^(-1)+琼脂9g·L^(-1),植株矮壮,茎粗,叶片厚,茎叶红,根系发,有利瓶苗的花芽分化. Techniques about in-vitro rapid propagation for miniaturization breeding of Lampranthustenuifolius were presented.Results show that the way that utilizes 0.1% mercuric chloride liquid to sterilize seeds of Lampranthustenuifolius for 5minutes is the best,and the survival rate of these seeds can reach 73.3%.Optimum culture medium for proliferation includes MS+6-BA 1.0mg·L^(-1)+ IBA0.05mg·L^(-1),sucrose 30g·L^(-1) and agar 9g·L^(-1).Additionally,growth of strong seedling illustrates that the best mass concentration of PP333 is 0.5mg·L^(-1).Culture medium for bud seedling contains MS+NAA 0.25mg·L^(-1),PP3330.5mg·L^(-1),sucrose 30g·L^(-1)and agar 9g·L^(-1).These plants are short but strong,and they have well developed root system,thick and red stems as well asleaves.It is beneficial to the in-vitro flower bud differentiation.
