摘要
为了获得具有较高酶活力的β-半乳糖苷酶,以乳糖为诱导剂,采用涂有x-gal的鉴别平板分离初筛得到28株生长较好的产β-半乳糖苷酶的菌株。通过摇瓶复筛测定其水解酶活,从中筛选出4株酶活力较高的菌株,并利用高效液相检测菌株的转糖基活性,确定了一株具有转糖苷活性菌株B5582Y,其初始水解酶活可达12 U/m L。根据细胞形态、生理生化数据、16S r DNA序列和rpo B功能基因序列数据多项鉴定分析,最终确定该菌B5582Y为克雷伯氏菌(Klebsiella michiganensis)。
In order to obtain high enzyme activity of β-galactosidase,twenty-eight strains which produced β-galactosidase were screened from soil using lactose as inducer and X-gal as indicator. Four strains which have higher hydrolysis enzyme activity were screened by shake flask culturing. One strain B5582Y which have both of hydrolysis and transgalactosylation Activity was screened by HPLC, its hydrolysis of initial enzyme activity can reach upwards of 12 U/mL. And it was identified as Klebsiella mich iganens is according to the cell morphology,physiological and biochemical characteristics, 16SrDNA and rpoB functional gene sequence.
