摘要
目的:研究新木脂素6-羟基-4-(4-羟基-3-甲氧基苯基)3-羟甲基-5-甲氧基-3,4-二氢(3R,4S)-2-醛基萘(VB-1)诱导人卵巢癌CoC1细胞凋亡作用。方法:体外培养CoC1细胞,碘化丙啶(PI)染色流式细胞术分析细胞凋亡率。比色法测定细胞Caspase-3活性。Western Blot分析Akt蛋白磷酸化水平。结果:VB-1显著增高CoC1细胞凋亡率和激活caspase-3(P<0.05),呈浓度依赖性。1.0μmol/L VB-1以时间依赖方式降低Akt蛋白磷酸化水平(P<0.05)。结论:VB-1能有效诱导CoC1细胞凋亡,其作用机制与抑制Akt蛋白磷酸化相关。
Objective To investigate whether 6-hydroxy-4-(4-hydroxy-3-methoxyphenyl)3-hydro-methyl-7-methoxy-3,4-di-hydro-2-naphthaldehyde(VB-1),a neolignan,induces apoptosis of human ovarian cancer CoC1 cells.Methods Human ovarian cancer CoC1 cells were cultured in vitro.The apoptotic rate was examined using flow cytometry using PI fluorescence staining.Caspase-3 activity was determined using colorimetric method.The level of the phosphorylated Akt protein expression was analyzed by Western Blot.Results VB-1 significantly induced increase of apoptotic cell death and activation of caspase-3 in CoC1 cells(P<0.01),in a concentration-dependent manner.VB-1(1.0μmol/L) a time-dependent reduced the level of the phosphorylated Akt protein expression(P<0.05).Conclusion VB-1 efficiently induces on apoptosis of human ovarian cancer CoC1 cells,and its mechanisms are involved in inhibition of Akt protein phosphorylation.
