长链非编码RNA VLDLR对人胰腺癌细胞增殖和迁移能力的影响

Effects of Long Non-Coding RNA VLDLR on Proliferation and Migration of Human Pancreatic Cancer Cell

摘要 该文探讨了长链非编码RNA(long non-coding RNA,1ncRNA)VLDLR在胰腺癌组织及5株细胞系中的表达,并分析了其对细胞增殖和迁移能力的影响。通过定量反转录PCR(quantitative reverse transcription PCR,qRT-PCR)检测VLDLR在14例胰腺癌组织和对应癌旁组织及5株细胞系中的表达水平,并进一步利用RNA干扰(ribonucleic acid interference,RNAi)技术探讨VLDLR的生物学功能。CCK-8和克隆形成实验检测细胞增殖能力,Transwell实验检测细胞迁移能力,Western blot检测蛋白质表达。结果发现,14例胰腺癌组织中,VLDLR相对表达水平为4.49±5.85(P<0.05),VLDLR在5株胰腺癌细胞中差异性表达。与对照组相比,实验组细胞中VLDLR m RNA水平明显降低(t分别为32.43、19.02,P<0.05);细胞增殖、细胞克隆形成及迁移能力明显均降低(P<0.05),基质金属蛋白酶-2(matrix metalloproteinase-2,MMP-2)和MMP-9蛋白质水平降低(P<0.05)。该研究结果表明,VLDLR在胰腺癌中高表达,下调VLDLR表达可降低胰腺癌细胞的增殖和迁移能力。 The aim of this paper is to investigate the expression profile of long non-coding RNA VLDLR in pancreatic cancer tissues and 5 different cell lines and the effect of VLDLR on the proliferation and migration of pancreatic cancer cell. Quantitative reverse transcription PCR(qRT-PCR) was performed to detect the relative expression of VLDLR in 14 cases of pancreatic cancer tissues and corresponding para-carcinoma tissues and five cell lines. The proliferation rates were examined by CCK-8 and colony formation assays. The migration rates were examined by Transwell assays. The relative level of the protein was detected by Western blot. The relative level of VLDLR m RNA in 14 cases of pancreatic cancer tissues was 4.49±5.85(P〈0.05). The relative level of VLDLR m RNA were different in five pancreatic cancer cell lines. Compared with the control group, VLDLR m RNA level was decreased(t=32.43, 19.02, P〈0.05) and the ability of proliferation, cell colony-forming and migration were significantly inhibited in experiment group(P〈0.05). The protein levels of matrix metalloproteinase-2(MMP-2) and MMP-9 were reduced(P〈0.05). These results indicated that the expression of VLDLR was up-regulated in pancreatic cancer. Knockdown of VLDLR could decrease the proliferation and migration of pancreatic cancer cell.
