半夏在动脉粥样硬化颈动脉血管内膜损伤中的修复作用研究 被引量:14

摘要 目的研究半夏对动脉粥样硬化大鼠颈动脉血管损伤修复中的作用及其机制。方法 6周龄SD大鼠随机分为5组,空白组、模型组、半夏治疗组,半夏、LY294002组、半夏+EPCs组,每组8只,除空白组外均建立颈动脉损伤模型,对各组损伤模型分别使用生理盐水、半夏水提液、半夏水提液+LY294002、半夏水提液+EPCs进行干预。苏木素伊红(HE)染色检测半夏对颈动脉损伤大鼠血管内膜增生的影响,免疫组化和western blot检测半夏对大鼠血管增生部位VEGF、p-VEGF、p-e NOS、e NOS表达的影响。结果 HE染色发现半夏能够降低颈动脉损伤小鼠血管新生内膜的增生。免疫组化和western blot检测同样发现半夏能够促进VEGF、p-VEGF、p-e NOS、e NOS表达。结论半夏水提液能够通过刺激血管修复因子的分泌和血管的再内皮化并抑制血管新生内膜的增生,从而在大鼠颈动脉损伤血管修复中发挥作用。 Objective To study the effect and mechanism of Pinellia ternate on the repair of carotid catheter injury model in atherosclerosis rats. Methods 6 - week - old SD rats were randomly divided into 5 groups, blank group, model group, Pinellia treatment group, Pinellia + LY294002 group, Pinellia + EPCs group, 8 rats of each group. In addition to blank group, the model of each group were treated with saline, Pinellia water extract, Pinellia water extract + LY294002, Pinellia water extract + EPCs intervention. The effect of Pinellia ternata on the proliferation of vascular endothelium in rats with carotid artery injury was detected by HE staining. Immunohistochemistry and western blot detected the expression of VEGF, p - VEGF, p - eNOS and eNOS. Results Western blot analysis found that Pinellia water extraction can up - regu-late the expression of protein such as VEGF, p - VEGF, p - eNOS and eNOS. HE staining analysis found that Pinellia water extraction can inhibit hyperplasia of carotid artery injury in mice rats with vascular neointimal. Conclusion Piniella has an effect on vascular endothelial function after intravascular injury in rats. Pinellia water extraction can stimulate the secretion of vascular repair factor and inhibit vascular intimal hyperplasia in the rat carotid artery injury model.
