摘要
目的:观察加减三甲散对肝纤维化大鼠肝脏微血管密度及PDGF-B表达的影响,从分子生物学角度探讨其作用机制。方法:选取SPF级Wistar雄性大鼠24只,随机分成正常组、模型组、加减三甲散组、阳性对照组(复方鳖甲软肝片)。采用猪血清腹腔注射法复制大鼠肝纤维化模型,以加减三甲散干预,并以复方鳖甲软肝片做对照药物,治疗60天后检测各组大鼠肝脏微血管密度(MVD)及PDGF-B的表达。结果:模型组大鼠MVD、PDGF-BmRNA及蛋白含量均较正常组升高(P<0.01),加减三甲散组大鼠MVD、PDGF-BmRNA及蛋白含量均较模型组下降(P<0.01或P<0.05),与鳖甲片组比较无差异(P>0.05)。结论:加减三甲散可在一定程度上逆转大鼠肝纤维化进程,其可能机制是通过下调大鼠肝组织中的PDGF-B的表达水平,减少肝脏病理性血管增生,降低微血管密度,从而抑制肝纤维化进程。
Objective:To investigate the influence of Modified Sanjia Powder on the microvessel density and the expression of PDGF-B in the liver of rats with hepatic fibrosis and explore its mechanism from the view of molecular biology.Methods:Twenty-four SPF Wistar male rats were selected and randomly divided into normal group,model group,Modified Sanjia Powder group,positive control group(Biejia Ruangan).Rat liver fibrosis model was induced by intraperitoneal injection of porcine serum,Modified Sanjia Powder intervention,and Fufang Biejia Ruangan as control drug.After 60 days of treatment,the microvessel density and PDGFB expression in liver of rats were detected.Results:Compared with normal group,the MVD and the content of PDGFB-mRNA and PDGFB protein of rats in model group increased(P〈0.01).Compared with model group,MVD and the content of PDGFB-mRNA and PDGFB of rats in the Modified Sanjia Powder group decreased(P〈0.01).All the above indicators had no significant difference between Modified Sanjia Powder and Fufang Biejia Ruangan group(P〉0.05).Conclusion:Modified Sanjia Powder could reverse the process of liver hepatic fibrosis in rats to a certain extent.Its possible mechanism is by down regulating the expression level of PDGF-B in liver tissue of rats,reducing pathological angiogenesis and microvessel density,so as to inhibit the development of liver fibrosis.
