摘要 目的优选青紫消斑颗粒中青黛、紫草、丹参的醇提工艺。方法以β,β-二甲基丙烯酰胺阿卡宁转移率为考察指标,采用L_9(3~4)正交试验优选紫草等醇提工艺。采用高效液相色谱法(high performance liquid chromatogra-phy,HPLC)测定阿卡宁含量,流动相乙腈∶水∶甲酸(70∶30∶0.05),检测波长:275 nm。结果最佳提取工艺为95%乙醇,40℃温浸2次,第1次加10倍量,第2次加8倍量,每次24 h。结论该工艺稳定可行,适用于青紫消斑颗粒的工业化生产。 Objective To optimize the alcohol extraction process of indigo naturalis, lithospermum and salvia miltiorrhiza. Methods Taken β,β- dimethylacrylalkannin transfer rate as the study index, L9(34) orthogonal test was usedfor optimization of extraction process with alcohol. Akanin content was tested by HPLC, the mobile phase was consisted of acetonitrile-water-formic acid(70:30: 0.05), at the wave lengths of 275 nm. Results The optimal ethanol extraction process was as following: dipping(40 ℃) extracting 2 times with the first time increased to 10 times, and 8 times for the second time, 24 hours for each time. Conclusion The optimized extraction process is stable, and suitable for the qingzixiaoban granules industrial production.
