不同质量浓度臭氧对家兔骨骼肌损伤修复过程TNF-α的影响 被引量:7

摘要 目的研究不同质量浓度的医用臭氧在家兔骨骼肌损伤部位局部注射对损伤组织及血浆中TNF-α的影响,评估其对骨骼肌急性损伤的疗效。方法将90只家兔随机分为空白组、模型组、臭氧治疗组(质量浓度分别为20,30和50μg·mL^(-1)的Ⅰ,Ⅱ和Ⅲ组)进行造模,术后12h开始治疗,分别在1,5和10d取材,检测损伤骨骼肌组织及血浆中TNF-α的变化。结果 TNF-α表达量:模型组与臭氧治疗Ⅰ组在1,5和10d均显著高于空白组;臭氧治疗Ⅱ和Ⅲ组在1和5d均显著高于空白组;臭氧治疗Ⅰ,Ⅱ和Ⅲ组在1,5和10d均显著低于模型组;臭氧治疗Ⅲ组在1和5d均显著低于臭氧治疗Ⅰ和Ⅱ组,在10d显著低于臭氧治疗Ⅰ组,与臭氧治疗Ⅱ组比较差异无统计学意义;臭氧治疗Ⅱ组在1,5和10d均显著低于臭氧治疗Ⅰ组。结论质量浓度为20,30和50μg·mL^(-1)的医用臭氧可显著降低家兔骨骼肌损伤部位与血浆中TNF-α的表达量,可减轻炎症、有效修复损伤的骨骼肌,其疗效随医用臭氧质量浓度的增加而增强。 Objective To study the effect of different concentrations of medical ozone on tissue and plasma TNF-αin rabbit skeletal muscle injury of local injection,and to assess the curative effect on the acute injury of skeletal muscle.Methods 90 modeling rabbits were randomly divided into control group,model group,ozone treatment groupⅠ(20μg·mL^-1),group Ⅱ(30μg·mL^-1),and group Ⅲ(50μg·mL^-1)then the treatment was implemented after 12 hof the operation.On the 1,5and 10 d,the change of TNF-αin damaged skeletal muscle tissue and in plasma was detected.Results The expression of TNF-αof the model group and the treatment group Ⅰ on 1,5and 10 dwere significantly higher than those of the blank group;meanwhile the treatment groupsⅡ,Ⅲ on1 and 5dwere significantly higher than those of the blank group.The expression of TNF-αof the treatment groupsⅠ,Ⅱ andⅢon 1,5and 10 dwere significantly lower than those of the model group.The expression of TNF-αof the treatment groupⅢ on 1and 5dwere significantly lower than those of the treatment groupsⅠandⅡ,but on 10 dwas significantly lower than those of the treatment group Ⅰ,and there was no statistical difference compared with the treatment groupⅡ.The expression of TNF-αof the treatment group Ⅱ on 1,5and 10 dwere significantly lower than those of the treatment groupⅠ.Conclusion The concentration of medical ozone at 20,30 and 50μg·mL^-1 can significantly reduce the expression of TNF-αin rabbit skeletal muscle injury and plasma,reduce inflammation,and can effectively repair the injury of skeletal muscle.The curative effect was increased with the increasing of ozone concentration.
