摘要
血管平滑肌细胞上大电导钙激活钾通道(BK_(Ca))在血管舒缩和血压调节中发挥了重要作用。其激活导致细胞膜超极化,从而引起血管平滑肌舒张。但在糖尿病或高糖状态下,BK_(Ca)功能受损,其对血管紧张性的调节作用降低,这可能是糖尿病患者心脑血管疾病发生的重要机制。近年来,泛素蛋白酶途径、肾素-血管紧张素系统信号通路对BK_(Ca)尤其是β_1亚单位的影响越来越受关注。因此,BK_(Ca)尤其是β_1亚单位及其调节途径中涉及的关键分子,有望成为治疗糖尿病引起的心脑血管病变的重要靶点。
Large conductance calcium-activated potassium channels(BK_(Ca)),abundantly expressed in vascular smooth muscle cells,play an important role in vasodilation and regulation of blood pressure. Activation of BK_(Ca)channels leads to the hyperpolarization of the plasma membrane,which eventually contributes to the vasodilation. Under the circumstance of diabetes or high blood glucose,BK_(Ca)channels are damaged,resulting in the losing of regulation function that the BK_(Ca)channels act on the vascular tone,which might be an important mechanism of cardiovascular and cerebrovascular diseases of diabetic patients. In recent years,the effects of ubiquitin-proteasome pathway and rennin-angiotensin system signal path on the BK_(Ca) channel,especially the β_1 subunit,have drawn more and more attention. Therefore,BK_(Ca)channels,especially the β_1 subunit,and the key molecules involved in the regulatory pathway,are expected to become the important targets to treat cardiac cerebrovascular diseases caused by diabetes.
