Evaluation of Functional Components of Lotus Leaves from Different Regions

摘要 测定广昌赣莲(江西广昌)、江苏红莲(江苏宝应)、洪湖莲(湖北洪湖)、湘白莲(湖南湘潭)、建莲(福建建宁)这5个产地的荷叶中生物碱、黄酮、甾醇及挥发物的含量。以乙醇水溶液为提取剂超声提取荷叶粉末,通过分光光度法分析,得到荷叶生物碱含量最高的是湘白莲4.08 mg/g;荷叶中黄酮含量最高的是宝应红莲19.99 mg/g。利用有机溶剂提取法得到甾醇粗提物,经过皂化,SPE柱萃取,衍生,氮吹后,GC-MS测定荷叶中甾醇含量。荷叶中甾醇含量最高的是宝应红莲0.25 mg/g,主要的甾醇为β-谷甾醇。利用水蒸气蒸馏法提取荷叶中的挥发油,并利用GC-MS测定挥发油的含量及成分,荷叶挥发油含量最高的是洪湖莲0.75%,荷叶挥发油中植醇、叶绿醇、烯类较多。 This research studied the contents of alkaloids, flavonoids, sterol as well as volatile oil from the lotus leaves of the five famous origins of lotus in China of Guangchang Lotus (Jiangxi Guangchang), Jiangsu Hong Lotus (Jiangsu Baoying), Hong Hu Lotus (Hubei Honghu), Xiang Lotus (Hu'nan) and Jian Lotus (Jianning, Fujian). With ethanol as extraction solvent , physiologically active substances was extracted from lotus leaves powder by ultrasonic extraction way, and measured with spectrophotometry. The highest content of alkaloids in lotus leaves was Xiang Lotus 4.08 mg/g. The highest content of flavonoids in lotus leaves was BaoYing Hong Lo- tus 19.99 mg/g. Ethanolic extract from lotus leaves was carried on with saponification, solid phase extraction, derivation, and nitrogen blow, and after that the content of sterol in lotus leaves was detected by Gas Chro matography-Mass Spectrometer. The highest content of sterol in lotus leaves was Bao Ying Hong Lotus 0.25 mg/g, and the main kind of sterol was β-sitosterol. The volatile oil was extracted by steam distillation, and its content was detected by Gas Chromatography-Mass Spectrometer. The highest content of volatile oil in lotus leaves was Hong Hu Lotus 0.75 %. The content of phytol and olefin played a major part in volatile oil of lotus leaves.
