摘要
目的应用miRNA芯片技术,研究血浆miRNA在鼻咽癌发生、发展中的表达差异性,初步筛选出与鼻咽癌相关的miRNA,为鼻咽癌早期发现、早期诊断、早期治疗提供依据。方法选取90例初次接受治疗的鼻咽癌患者及健康成年人60例,分别收集所有研究对象的血浆标本,分离miRNA,运用Gene Pix4000B芯片行比较分析病例组与对照组之间miRNA的表达是否存在差异。结果通过比较,各组一般资料无差异,90例鼻咽癌患者临床分期为Ⅰ期组20例,Ⅱ期组20例,Ⅲ期组25例,Ⅳ期组25例;通过miRNA芯片检测发现差异表达miRNA31个,在这31个miRNA中,上调有26个,下调有5个,差异具有统计学意义(P<0.05)。结论鼻咽癌患者和健康人血浆中含有的miRNA表达存在差异,提示miRNA参与到鼻咽癌病情的发生以及发展当中,可为鼻咽癌的早期发现,早期诊断,早期治疗提供理论依据。
Objective To study the differential expression of plasma miRNA in the occurrence and development of nasopharyngeal carcinoma by miRAN chip technology,to conduct the preliminary screening of the miRNA associated with nasopharyngeal carcinoma,and to provide evidence for early detection,early diagnosis and early treatment of nasopharyngeal carcinoma. Methods 90 patients with nasopharyngeal carcinoma in the initial treatment and 60 cases of healthy adults were selected.Plasma samples were collected from all subjects to separate the miRNA.GenePix4000B chip was used to compare and analyze the differential expression of case group and control group. Results By comparison,there was no difference in general data.90 patients with nasopharyngeal carcinoma were divided into stage I group(n=20),stage II Group(n=20),stage III group(n=25)and stage IV group(n=25).There were 31 cases different in the expression of miRNA by miRNA.Among the 31 cases of miRNA,there were Up by 26,down by 5,and the difference was statistically significant(P 〈 0.05). Conclusion The expression of miRNA in plasma of NPC patients and healthy people is different.It is suggested that miRNA is involved in the occurrence and development of NPC.It can provide theoretical basis for early detection,early diagnosis and early treatment of nasopharyngeal carcinoma.
