摘要
目的探索和改进小鼠脓毒症模型构建方法;方法 120只雄性小鼠随机分为12组,实验分四组,每组设置三个平行组,采用直径2mm三棱针予盲肠结扎和穿孔,A组(距离盲肠末端1/3处结扎,穿刺1孔),B组(距离盲肠末端1/3处结扎,穿刺2孔),C组(距离盲肠末端2/3处结扎,穿刺1孔),D组(距离盲肠末端2/3处结扎,穿刺2孔),观察72h死亡情况;结果 A组死亡率0%~50%,B组死亡率30%~50%,C组死亡率30%~60%,D组死亡率80%~90%;结论研究药物对脓毒症的治疗作用建议用B组建模,而观察药物保护实验则建议选用D组。
OBJECTIVE To explore and improve the method for establishing the mice model of sep-sis. METHODS We divided 120 male mice into 12 groups randomly, which were divided into four big groups(group A, group B, group C and group D) and each group sets up three parallel groups. Then the cecal ligation andperforation(CLP) surgery was performed by three-edged needle with the diameter of 2ram. Group A( ligated at 1/3end of cecum and punctured 1 hole), group B (ligated at the 1/3 end of cecum and punctured 2 hole), group C (li-gated at the 2/3 end of cecum and punctured 1 hole), group D ( ligated at the 2/3 end of cecum and punctured 2 hole). The mortality rate of mice were observed until 72 hours. RESULTS The mortality rate of mice range from 0% to 50% in group A, from 30% to 50% in group B, from 30% to 60% in group C, and from 80% to 90% in group D. CONCLUSION Group B was recommended for researching the therapeutic effect of drugs on the sepsis, while group D was recommended for the drug protection experiments.
