摘要
为研究桦褐孔菌菌质多糖对小鼠脾淋巴细胞增殖的影响,采用水提醇沉法提取粗多糖,Sevag法脱蛋白后透析,再经DEAE-Sepharose CL-6B离子交换柱进一步分离纯化,MTT法检测不同浓度的桦褐孔菌菌质多糖对小鼠脾淋巴细胞增殖的影响。试验结果得到3个多糖纯化组分JZP1、JZP2、JZP3;粗多糖(JZPC)、精制多糖(JZPJ)和纯化多糖(JZP3),能够促进淋巴细胞增殖,最大增殖率分别为59.04%(1 000μg/m L),44.58%(20μg/m L),39.76%(20μg/m L)。JZP1和JZP2在1 000μg/m L时抑制淋巴细胞增殖,抑制率分别15.66%和13.25%。结果表明相同浓度下粗多糖对小鼠脾淋巴细胞的增殖作用强于纯化多糖,不同多糖组分表现出不同的免疫学活性,并且其免疫活性与其浓度密切相关。
In order to study the influence of polysaccharides from Inonotus obliquus Bi-direction Solid Fermentation on proliferation function of mouse splenic lymphocyte, crude polysaccharides were extracted using boiling ethanol precipitation method, and purified by Sevag, dialysis and DEAE-Sepharose CL-6B chromatography. The effect of polysaccharides on mice splenic lymphocytes was analyzed by MTT method. Three purified polysaccharides JZP1 ,JZP2,JZP3 were obtained in this experiment. These polysaccharides JZPC, JZPJ, JZP3 could significantly improve lymphocytes proliferation, the maximum proliferation rate was 59.04%( 1 000 μg/mL) ,44.58%(20 μg/mL), 39.76%(20 μg/mL), respectively. JZP1 and JZP2 at the concentrations of 1 000 ixg/mL significantly inhibited lymphocytes proliferation, the inhibition rate was 15.66%and 13.25%, respectively. At the same concentration, the effect of crude polysaceharide was stronger than that of the purified polysaeeharide on the proliferation of spleen lymphocytes in mice.The different polysaceharide fractions exhibited different immunological activity, and its immunological activity was closely related to its concentration.
