摘要
目的建立镍变应性接触性皮炎(allergic contact dermatitis,ACD)小鼠模型。方法选择30只C57BL/6小鼠,随机分为5组(阳性对照组,阴性对照组,5%-5%硫酸镍组,10%-10%硫酸镍组,10%硫酸镍单次刺激组),于第0天致敏且第2、3天激发,分别于第一次激发后的6、12、24、48 h对各组激发部位炎症进行动态评分;超声扫描表皮厚度;苏木精-伊红染剂(hematoxylin and eosin stain,HE)染色观察激发部位皮肤病理变化;对皮损部位皮肤匀浆进行细胞因子检测。结果各剂量组动物激发部位皮肤24 h炎症程度最强。各剂量组激发部位皮肤表皮增厚,超声扫描显示实验组小鼠表皮厚度为:10%-10%硫酸镍组>5%-5%硫酸镍组>10%硫酸镍单次刺激组。5%-5%、10%-10%硫酸镍组出现表皮结痂,基底层液化,真皮毛细血管扩张,真皮炎性细胞浸润,表现为急性炎症症状,与人类镍过敏的急性病理变化一致。5%-5%、10%-10%硫酸镍组皮损部位干扰素-γ、白细胞介素-4和白细胞介素-10均值水平均有升高,10%-10%硫酸镍组的白细胞介素-4均值水平与阴性对照组相比,差异存在统计学意义(P<0.05)。结论 5%-5%、10%-10%硫酸镍组成功建立ACD急性小鼠模型。
Objective To establish a nickel-induced allergic contact dermatitis (ACD) mouse model. Methods Thirty C57BL/6 mice were divided into 5 groups based on the concentration of nickel sulfate (5%-5%, 10%-10%, 10% single administration, positive control and negative control). All mice were sensitized at day 0 and challenged at day 2 and day 3. Grading of the skin alterations was performed at 6, 12, 24, and 48 h after first challenge respectively. The skin thickness of mice was detected by ultra- sonic scan. Pathological changes were evaluated by hematoxylin and eosin stain (HE) staining among cu- taneous biopsy samples. IFN-γ, IL-4, and IL- 10 levels were detected in skin homogenates 48 h after challenge. Results The severest inflammatory reactions were observed at 24 h in all nickel sulfate chal- lenge groups and epidermis incrassation were observed in all nickel sulfate challenge groups. The thickness of epidermis based on the results of ultrasonic scan were as follow: 10%-10%〉5%-5%〉10% single challenge group. Acute inflammatory reaction such as escharosis on epidermis, liquefaction of stra-tum basalis, angiotelectasis and inflammatory cell infiltration in dermis was observed in 5%-5% and 10%- 10% group, which is similar to pathological feature of acute phase of human ACD. The cutaneous levels of interferon-,/, interleukin-2, interleukin-10 of 5%-5% and 10%-10% group are higher than negative con- trol group, the differences were significant statistically (P〈0. 05). Conclusion The mouse models of nickel-induced ACD had been established successfully in 5%-5% and 10%-10% group.
