摘要
目的:探讨PI3K/Akt抑制剂NVP-BEZ235对血管肉瘤细胞系ISOHAS增殖及侵袭的影响。方法:使用CCK8检测24h、48h、72h不同浓度NVP-BEZ235(5、10、20、50、100nM)处理后ISOHAS细胞的增殖能力。Transwell小室实验、Western Blot实验检测NVP-BEZ235(50、100n M)处理后ISOHAS细胞的侵袭能力及细胞内PI3K/Akt/mTOR信号通路相关蛋白表达变化情况。结果:与对照组比较,NVP-BEZ235处理后血管肉瘤细胞的增殖、侵袭能力明显下降,且细胞中PI3K/Akt/mTOR信号通路相关蛋白p-Akt、p-mTOR的表达明显下降。结论:NVP-BEZ235通过阻断PI3K/Akt/mTOR信号通路抑制血管肉瘤细胞的增殖、侵袭能力,提示NVP-BEZ235对血管肉瘤治疗具有潜在的应用价值。
Objective: To explore the role of NVP-BEZ235 on the proliferation and invasion of angiosareorna cells. Methods: Culture the ISOHAS in vitro, assess the different doses (0, 5, 10, 20, 50, 100 nM) NVP-BEZ235 inhibition effection on proliferation of ISOHAS by CCK8 after 24h, 48h, 72h. The invasion ability of ISOHAS cells were evaluated by transwell chamber assay.Western blot was performed to detect the expression of PI3K/AKT/mTOR signaling pathway related proteins.Results:Compared with the control group, after NVP-BEZ235 treatment, the proliferation and invasion ability of angiosarcoma cells decreased significantly, and the expression of PI3K/Akt/mTOR signaling pathway in cell associated proteins p-Akt and p-roTOR decreased. Conclusion:NVP-BEZ235 inhibits the proliferation and invasion ability of angiosareoma cells by blocking PI3K/Akt/mTOR signaling, it suggested that NVP-BEZ235 has potential applications in the treatment of angiosarcoma.
